摘 要:目的:观察麦冬多糖对2型糖尿病围绝经期模型大鼠血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)和丙二醛(MDA)水平的影响,并探讨其作用机制。方法:大鼠予高糖、高脂饲料喂养8周后,采用链脲佐菌素诱导联合切除两侧卵巢的方法建立2型糖尿病围绝经期大鼠模型。选取50只建模成功的雌性大鼠随机分为模型组、阳性对照组(造模后第10天予二甲双胍0.25 g·kg^-1)以及麦冬多糖低、中、高剂量组(造模后第10天分别予100、200、400 mg·kg^-1麦冬多糖),每组10只。另取10只大鼠作为假手术组。比较治疗前后各组大鼠体质量、血糖水平、SOD、GSH-Px、CAT和MDA水平的差异。ELISA法检测各组大鼠血清雌二醇(E2)、促卵泡生成素(FSH)和促黄体生成素(LH)水平的差异。结果:与模型组比较,阳性对照组以及麦冬多糖低、中和高剂量组大鼠体质量明显增加,血糖水平明显降低(均P<0.05),血清SOD、GSH-Px和CAT水平明显升高(均P<0.05),血清MDA水平明显降低(P<0.05),血清E2水平明显升高(P<0.05),血清FSH和LH水平明显降低(均P<0.05)。结论:麦冬多糖对2型糖尿病围绝经期有一定保护作用,其机制可能与提高抗氧化能力和调节血清雌激素水平有关。Objective:To observe the effects of Ophiopogon japonicuson polysaccharide on serum superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and malondialdehyde(MDA)in diabetic peri-menopausal rats,and explore its mechanism of action.Methods:The type 2 diabetes premenopausal rats were established by combined removal of bilateral ovaries with streptozotocin induction after giving high sugar and high fat feed for 8 weeks.Fifty model rats were randomly divided into model group,positive control group(melbine 0.25 g·kg^-1 was given 10 days after molding)and Ophiopogon japonicuson polysaccharide low,medium and high dose group(100,200,400 mg·kg^-1 were given respectively 10 days after molding),10 in each group.Another 10 rats were selected as the sham operation group.The levels of body weight,blood glucose,serum SOD,GSH-Px,CAT and MDA in each group were compared before and after treatment.Levels of serum estradiol(E2),follicle-stimulating hormone(FSH)and luteinizing hormone(LH)in each group were detected by ELISA and compared each other.Results:Compared with the model group,the body weight of the rats in the positive control group and low,medium,high dose polysaccharide groups were increased and blood glucose levels were decreased significantly(all P<0.05).The levels of serum SOD,GSH-Px and CAT were significantly increased(all P<0.05),and serum MDA were significantly decreased(P<0.05);serum E2 was significantly increased(P<0.05),serum FSH and LH were significantly decreased(all P<0.05).Conclusion:Ophiopogon japonicuson polysaccharide can protect type 2 diabetes perimenopenesis,and its mechanism may be related to improving antioxidant capacity and regulating estrogen levels.
