消痞颗粒对胃癌前病变大鼠p16基因甲基化的干预研究

摘　　要：目的探讨消痞颗粒对胃癌前病变大鼠p16甲基化的干预机制。方法60只大鼠随机分为空白组10只,模型组50只,模型组以N-甲基-N′-硝基-N-亚硝基(MNNG)溶液灌胃为主要负荷因素建立胃癌前病变大鼠模型。造模后,将造模组大鼠按照体质量随机分为盐水组、维酶素组、消痞颗粒组。盐水组给予生理盐水灌胃;维酶素组予维酶素混悬液灌胃;消痞颗粒组予消痞颗粒混悬液灌胃,干预持续12周。选用甲基化特异性PCR法(MSP)、免疫组化检测大鼠p16启动子CpG岛甲基化表达水平及其蛋白转录表达水平。结果消痞颗粒可能通过降低p16CpG岛甲基化水平进而上调其蛋白表达水平,从而达到阻断胃癌发生发展。结论消痞颗粒通过干预抑癌基因p16甲基化及蛋白表达水平有效逆转胃癌前病变。Objective To investigate the interventional mechanism of Xiaopi Granules in p16 methylation of rats with precancerous lesions of gastric cancer.Methods 60 rats were randomized into the blank group(n=10)and the model groups(n=50).Rats in the model groups were given intragastric administration with N-methyl-N′-nitro-N-nitrosoguanidine(MNNG)to establish the precancerous lesions model.According to the weight sequence,the model rats were randomly divided into saline group,vitacoenzyme group,Xiaopi Granules group,which were given gavage with saline,vitacoenzyme,Xiaopi Granules respectively for 12 weeks.Methylation-Specific PCR(MSP)and immunohistochemical staining(IHC)were applied to detect expression of p16 promoter CpG island methylation and the mRNA expression of protein.Results Xiaopi Granules decreased p16 CpG methylation and up-regulated the expression of protein to inhibit the development of gastric cancer.Conclusion Xiaopi Granules can effectively treat precancerous lesions of gastric cancer by intervening p16 methylation and protein expression.
