摘 要:目的探讨中药复方凉血固元汤对大鼠急性放射性肠损伤的作用机制。方法将30只雌性SD大鼠随机分为空白组、模型组、地塞米松组及凉血固元汤高、低剂量组5组。除空白组外,其余各组采用60 Co-γ射线单次全身照射建立放射性肠损伤模型,自照射后第1天至第3天,各组给予相应药物灌胃。空白组、模型组给予生理盐水,地塞米松组给予地塞米松溶液(1.4 mg/kg),凉血固元汤高、低剂量组分别给予36.0 g/kg、9.0 g/kg凉血固元汤溶液灌胃,连续3 d。每日观察记录大鼠腹泻、死亡率、进食量等一般情况,在照射后第3天,麻醉并处死各组大鼠,取小肠组织进行免疫组化检测,比较各组TLR4、MyD88、NF-κB表达差异。结果各照射组大鼠均出现不同程度的腹泻,模型组为水样便,各治疗组相对较轻;各照射组大鼠体质量明显下降,第3天凉血固元汤高剂量组降幅最小。各组TLR4表达比较:模型组较空白组明显升高(P<0.01),地塞米松组和凉血固元汤高、低剂量组表达量均降低(P<0.05);各组MyD88比较:模型组、凉血固元汤低剂量组较空白组明显升高(P<0.01,P<0.05),地塞米松组、凉血固元汤高剂量组均降低(P<0.05);各组NF-κB比较:模型组NF-κB表达高于空白组(P<0.05),凉血固元汤高剂量组低于模型组(P<0.05)。结论凉血固元汤可有效减轻由电离辐射引发的放射性肠损伤,改善大鼠生存状态,其作用机制可能与抑制TLR4/NF-κB通路相关因子表达有关。Objective To investigate the mechanism of Liangxue Guyuan Decoction(LGD)on acute radiation-induced intestinal injury in rats.Methods 30 female SD rats were randomly divided into 5 groups:blank group,model group,dexamethasone group,low and high dose LGD group.Except the blank group,the rats were irradiated by 60Co-γray(single total body)to build models.From day 1 to day 3 after irradiation,each group was given gastric administration for 3 days.The blank group and the model group were given saline;the dexamethasone group was given dexamethasone solution(1.4 mg/kg);the low and high dose LGD groups were given 36.0 g/kg,9.0 g/kg LGD respectively for 3 days.General conditions such as diarrhea,death rate and food intake were observed daily.On the third day after irradiation,rats in each group were anesthetized and sacrificed.The expression of TLR4,MyD88 and NF-κB in small intestine tissues was detected by immunohistochemistry.Results After irradiation,rats in all irradiation groups had different degrees of diarrhea.The model group had watery stools,which were relatively mild in each treatment group.The weight of rats in each irradiation group decreased significantly,and the high dose LGD group had the smallest decrease on the third day.Compared with the blank group,TLR4 expression in the model group was significantly higher(P<0.01),while the expression in the dexamethasone group,the low and the high dose LGD groups decreased(all P<0.05);MyD88 expression in the model group and the low dose LGD group was significantly higher than the blank group(P<0.01,P<0.05),while the expression of the dexamethasone group and high dose LGD group decreased(P<0.05).the expression of NF-κB in the model group was higher than the blank group(P<0.05),and the expression in the high dose LGD group was lower than that in the model group(P<0.05).Conclusion Liangxue Guyuan Decoction could effectively alleviate the radiation-induced intestinal injury and improve the survival status of rats.Its mechanism may be related to the inhibition of the ex
