摘 要:以苏云金杆菌和韭菜迟眼蕈蚊科为试材,采用浸泡韭菜茎段法,设计了5个韭菜茎段浸泡时间、5个苏云金杆菌稀释浓度和4个处理观察时间,以期确定苏云金杆菌对韭菜迟眼蕈蚊幼虫室内毒力测定时的最佳韭菜茎段浸泡时间、苏云金杆菌稀释浓度和处理时间。结果表明:室内测定苏云金杆菌毒力时,以韭菜茎段的浸泡时间60 s、苏云金杆菌稀释浓度500 mg·L-1、处理观察时间24 h为宜,该条件下试虫平均死亡率为90%。通过计算不同处理时间苏云金杆菌不同稀释浓度的毒力方程,确定处理观察24 h时半致死浓度为49.40 mg·L-1。Bacillus thurinensis and Bradysia ordoriphaga were used as experimental materials.With the method of soaking chives stem sections,five soaking time of chives stem sections,five diluted concentration of B.thuringiensis and four observation time after treatment were designed in the experiment.The experiment purpose was to give the best soaking time,diluted concentration and observation time when indoor toxicity of B.thurinensis to B.ordoriphaga larva was tested.The results showed by difference significance analysis,that the optimal test factors soaking time of leek stem sections was 60 seconds,diluted concentration of B.thuringiensis was 500 mg·L-1,observation time after treatment was 24 hours.Under the above experimental conditions,the average mortality of B.thuringiensis larva was 90%.Toxicity equations of diluted concentration of B.thuringiensis were simulated at different observation time after treatment.When the observation time after treatment was 12 hours,semi-lethal concentration LC50 was 49.40 mg·L-1.
