摘 要:目的探讨软脉煎抗动脉粥样硬化作用在代谢物水平的调控机制。方法收集经复方软脉煎治疗前后的肾精亏虚型颈动脉粥样硬化患者,应用超高压液相色谱-电喷雾-四级杆飞行时间串联质谱分析法对样本进行全谱分析,采用XCMS对数据进行峰提取和代谢物鉴定,运用主成分分析、判别分析等统计方法筛选肾精亏虚型颈动脉粥样硬化患者经软脉煎治疗前后的差异代谢物,并进行相关通路分析。结果正、负离子模式下,肾精亏虚型颈动脉粥样硬化患者经软脉煎治疗后与治疗前比较,具有显著性差异的代谢物共5种:次黄嘌呤、胆酸、腺苷酸、腺嘌呤、乙酰-DL-亮氨酸;所有差异代谢物共参与64条通路。结论软脉煎可能通过干预与次黄嘌呤、胆酸、腺苷酸、腺嘌呤、乙酰-DL-亮氨酸等代谢物相关的通路发挥抗动脉粥样硬化作用。Objective To explore the regulatory mechanism on metabolite levels of ruanmai decoction on carotid atherosclerosis CAS.Methods Plasma samples of patients of type of kidney essence deficiency with carotid atherosclerosis before and after the treatment of compound ruanmai decoction were collected and divided into three groups. UHPLC-Q-TOF MS was carried out to analysis the full spectrum of the samples, XCMS was used to extract the peak and identify the metabolites. Principal component analysis and discriminant analysis were used to screening differences metabolites between patients of type of kidney essence deficiency with CAS before and after the treatment of ruanmai decotion. Results In positive and negative ion mode, there were 5 kinds of significantly different metabolites in the patients of type of kidney essence deficiency with carotid atherosclerosis before and after the treatment of ruanmai decoction, including hypoxanthine, hypoxanthine, adenosine, adenosine and acetyl-DL-leucine. All of the significantly metabolites were participated in the 64 metabolic pathways. Conclusion Ruanmai decoction seems to anti-atherosclerosis by intervening in pathways related to the significant metabolites such as hypoxanthine, hypoxanthine, adenosine, adenosine and acetyl-DL-leucine.
