摘  要:低水平光疗法(LLLT)是治疗皮肤病的常用方法。目前,关于脉冲光照射是否优于连续光照射模式及其原因仍有待讨论。本文设计制作了脉冲黄光LED装置,使其在各种占空比下保持恒定的平均辐射照度。使用该装置比较了连续和脉冲光照对皮肤细胞黑素体成熟和黑色素合成的影响。实验分别检测了主要的生物学评估指标,包括TYR/TRP-1/MITF的细胞活力、酪氨酸酶活性,以及mRNA和蛋白质表达。实验结果表明,连续和脉冲LED光照均可抑制黑色素合成,其特征是TYR和TRP-1的mRNA和蛋白水平均显著降低。与连续模式相比,脉冲光模式对黑色素合成具有更好的抑制作用。Low-level light therapy (LLLT) is a popular method for skin disorders treatment. Whether and why pulsed light delivery is superior to continuous mode remains to be discussed. A pulsed yellow light-emitting diode (LED) device is proposed, which can keep constant average irradiance at various duty cycle. The effect of continuous and pulsed light delivery on melanosomes maturation and melanin synthesis has been compared by assessing skin cell viability, tyrosinase activity, mRNA and protein expression of TYR/TRP-1/MITF which are the three critical melanogenic enzymes in melanin synthesis. The results show that treatment with continuous and pulsed LED can inhibit melanin synthesis, characterized by decreased TYR and TRP-1 expression on both mRNA and protein levels. Pulsed light delivery has better inhibiting effect on melanin synthesis with lower TYR and TRP-1 expression compared to continuous mode.
