摘  要:以大黄花虾脊兰自然结实120~150d成熟未开裂的蒴果作为研究材料,对56个果荚中种子的有胚率进行统计,并采用TTC法测定种子活力,选取最佳用于无菌播种的果荚.以活力最佳的果荚为研究材料,对在不同含量的NAA、6 BA、KT及不同有机添加物下种子萌发及幼苗生长情况进行观察.结果表明:大黄花虾脊兰种子的有胚率为15.57%~95.66%,平均值为45.15%±2.68%,授粉后140 d的蒴果种子活性最佳;种子萌发120和140 d后种胚突破种皮生长为原球茎,150 d顶端分生出明显的芽生长点,底部生出肉质的不定根;较佳的培养基组合为:2 g·L^-1 Hyponex 1+1/2 MS+0.37 mg·L^-1 MgSO 47H2O+25 g·L^-1蔗糖+1.0 mg·L^-16 BA+0.2 mg·L^-1 NAA+1.0 mg·L^-1 KT+5.5 g·L^-1琼脂粉+1.5 g·L^-1活性炭+2 g·L^-1牛肉粉蛋白胨+50g·L^-1洋葱,pH5.7~5.8,培养(140±10)d可形成原球茎,萌发率达39.64%±3.34%.To develop a scale cultivation model for Calanthe striata R.Br.,120-150 days old mature and uncracked C.striata capsule obtained by natural seed setting were selected as the test material.Embryogenic rate of seeds from 56 pods was counted,and seed vigor was determined by TTC method to select the most vigorous seed for aseptic seeding.Then seed germination and seedling growth tests were conducted upon the pod with the best vitality under different concentrations of NAA,6BA,KT and organic additives using L9 orthogonal design.The results showed that the percentage of embryo in C.striata seeds ranged between 15.57%and 95.66%,averaging at 45.15%±2.68%.Capsule on the 140th day after seed set had the highest vitality.After 120 or 140 d,the embryo broke through the seed coat and grew into protocorm.After 150 d,obvious bud growing points were developed at the apical tissue,and fleshy adventitious roots were produced at the bottom.A preferred medium is the combination of 2 g·L^-1 Hyponex 1,1/2 MS,0.37 mg·L^-1 MgSO4·7H2O,25 g·L^-1 sucrose,1.0 mg·L^-16BA,0.2 mg·L^-1 NAA,1.0 mg·L^-1 KT,5.5 g·L^-1 agar powder,1.5 g·L^-1 activated carbon,2 g·L^-1 beef powder peptone,50 g·L^-1 onion under 5.7-5.8 pH.Protocorm was developed(140±10)d after cultivation,with the germination rate being 39.64%±3.34%.
