摘  要:采用DNA电泳、qRT-PCR及Western blot技术分析了在无氮和含氮培养条件下,巨菌草内生固氮菌Klebsiella variicola GN02菌株nifH基因序列长度、基因与蛋白表达量的差异.结果表明,GN02菌株在无氮及含氮条件下均有nifH基因表达,其克隆得到的nifH基因与同属禾本科的甘蔗的nifH基因亲缘关系较近;无氮培养条件下nifH基因和蛋白表达量明显高于含氮条件下的表达量.表明无氮条件能诱导GN02菌株nifH基因表达量的增加,而氮源抑制nifH基因的表达.To investigate the effect of nitrogen on the nifH gene expression of Klebsiella variicola GN02,an endophytic nitrogen-fixing bacteria isolated from Pennisetum giganteum,DNA electrophoresis,qRT-PCR and Western blot were used to analyze the sequence length of the gene,and differential expressions of gene and proteins of nifH in nitrogenous and nitrogen-free cultures.The results showed that the nifH gene of GN02 strain were both expressed in cultures with divergent nitrogen contents,and the genetic relationship of nifH gene cloned from GN02 strain was close to that of sugarcane,which is also a gramineous plant like P.giganteum.The expressions of nifH gene and protein of GN02 strain in nitrogen-free culture was significantly higher than that of nitrogenous culture.Therefore,nitrogen-free condition is likely to induce the expression of nifH gene in GN02 strain and nitrogen source tends to suppress the expression.
