摘  要:目的探讨蝙蝠葛碱(Dau)对BxPC-3的增殖和迁移作用,以及通过观察Dau对BxPC-3细胞ERK信号通路中Raf和MEK1 mRNA及蛋白表达的影响,进一步探讨Dau抗胰腺癌的作用机制。方法将BxPC-3细胞进行体外培养,以不同浓度的Dau进行药物干预,采用细胞划痕实验观察BxPC-3细胞的迁移能力;MTT法检测BxPC-3细胞增殖的抑制作用;Real-time PCR法检测BxPC-3细胞中Raf mRNA和MEK1 mRNA的表达情况;Western blot法检测BxPC-3细胞中Raf蛋白和MEK1蛋白的表达情况。结果Dau能够明显降低BxPC-3细胞的运动能力(P<0.01)。Dau高、低剂量组和5-FU组增殖抑制率分别为70.81%、44.98%、58.50%。Dau能下调BxPC-3细胞中Raf mRNA和MEK1 mRNA的表达,且差异显著(P<0.01)。下调BxPC-3细胞中Raf蛋白和MEK1蛋白的表达,且差异显著(P<0.05或P<0.01)。结论Dau能够明显抑制胰腺癌BxPC-3细胞的增殖和运动能力,明显抑制ERK信号通路关键位点Raf、MEK1 mRNA及蛋白的表达。Objective To explore the effect of Dau on the proliferation and migration of BXPC-3 cells,and the mechanism of Dau against pancreatic cancer by observing the influence of Dau on the expression of RAF and MEK1 in ERK signaling pathway.Methods Pancreatic cancer BXPC-3 cells were cultured in vitro and treated with Dau at different concentrations.Cell wound scratch assay was used to determine the migration ability of BxPC-3 cells.MTT experiment was employed to measure the proliferative inhibition of BxPC-3 cells.Real-time PCR was adopted to analyze the expression of Raf and MEK1 mRNA.Western blot was employed to test the expression of Raf and MEK1 protein.Results Dau remarkably reduced the migration of BxPC-3 cells(P<0.01).The cell proliferation inhibition rates were 70.81%,44.98%and 58.50%in high and middle dosages of Dau groups and 5-FU group,respectively.Dau reduced the expression of Raf and MEK1 mRNA and protein.Conclusion Dau could remarkably restrain the proliferation and migration of BxPC-3 cells and suppress the expression of Raf and MEK1 in ERK signaling pathway.
