摘  要:目的研究安五脂素对D-半乳糖致小鼠心肌损伤的保护作用。方法将雄性ICR小鼠随机分为5组,即空白对照组(CON),衰老模型组(D-gal)和低、中、高三个剂量的安五脂素给药组(AN-L、AN-M、AN-H),连续给药7周。造模成功后,使用试剂盒测定各组小鼠心肌组织中超氧化物歧化酶(SOD)、丙二醛(MDA)水平,通过免疫蛋白印迹法测定心肌组织Nrf2、血红素氧合酶1(HO-1)水平。结果与D-gal组相比,AN-M、AN-H显著升高小鼠心肌组织SOD活性(P<0.05或P<0.01),显著降低MDA含量(P<0.05或P<0.01)。与CON组相比,D-gal模型组Nrf2(P>0.05)、HO-1表达升高(P<0.05);与D-gal组相比,AN-H的Nrf2及HO-1表达显著升高(P<0.01,P<0.05)。结论安五脂素通过其抗氧化作用对D-半乳糖所致衰老小鼠心肌损伤具有显著的保护作用。Objective To study the protective effect of Anwulignan on myocardial injury induced by D-galactose in mice.Methods Male ICR mice were randomly divided into five groups:the blank control group(CON),the aging model group(D-gal),and the low,medium,and high doses of Anwulignan administration groups(AN-L,AN-M,AN-H),and treated respectively for consecutive 7 weeks.After successful modeling,the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in the myocardialtissue of each group were determined with kits.The levels of Nrf2 and HO-1 in myocardial tissue were determined by immunoblotting.Results Compared with D-gal group,AN-M and AN-H significantly increased SOD activity(P<0.05 or P<0.01)and significantly decreased MDA content(P<0.05 or P<0.01)in mouse myocardial tissue.Compared with the CON group,Nrf2(P>0.05)and HO-1(P<0.05)expression were increased in the D-gal model group.Compared with the D-gal group,the expression of Nrf2 and HO-1 in AN-H were significantly increased(P<0.01,P<0.05).Conclusion Anwulignan exerted a significant protective effect on D-galactose-induced myocardial injury in aging mice.
