摘 要:目的:探讨刘延庆教授临床治疗痛风的经验方对于高尿酸血症大鼠模型相关免疫炎症因子的影响,为该复方的临床应用及后期开发提供实验依据。方法:复方水煎剂在扬州市中医院中药制剂室制备。高尿酸血症模型造模通过给大鼠喂食定制高酵母饲料和腺嘌呤溶液,同时皮下注射氧嗪酸钾悬液进行造模。模型组大鼠分为阴性对照组(不给药)、给药组以及阳性对照组(秋水仙碱),连续给药10 d。全自动生化分析仪用来检测血尿酸水平,蛋白热荧光芯片用来检测痛风相关的免疫炎症因子的表达情况。显微镜下观察大鼠肝肾组织是否损伤。结果:与阴性对照组比较,该复方能明显降低高尿酸血症大鼠模型的血尿酸水平(P<0.01);与对照组相比,该复方对痛风相关的免疫炎症因子IFN-γ、IL-1β、IL-2、IL-4、IL-13、TNF-α等具有明显的抑制作用。显微镜下观察大鼠的肝肾组织,未发现明显的肝肾组织损伤。结论:该复方可以明显降低高尿酸血症大鼠模型的尿酸水平,同时抑制痛风相关免疫炎症因子,短期内并未发现明显的肝肾损害。这说明该复方具有明显的抗炎免疫作用,对于高尿酸血症具有明显治疗作用且无毒副作用。Objective:To explore the effect of Chinese herbal compound(Professor Liu Yanqing′s experience prescription)on immune-inflammatory factors in hyperuricemia rats.Methods:The decoction of compound was prepared in the Chinese medicine preparation room of Yangzhou Hospital of Traditional Chinese Medicine.The model of hyperuricemia was established by feeding rats with high yeast feed and adenine solution and injecting potassium oxalate suspension subcutaneously.Rats in model group were divided into negative control group(without administration),administration group and positive control group(colchicine),and were treated for 10 days continuously.Automated biochemical analyzer was used to detect the level of serum uric acid,and protein thermo-fluorescence chip was used to detect the expression of gout-related immune-inflammatory factors.At the same time,HE staining was performed to observe whether the liver and kidney tissues of rats were damaged.Results:Compared with the negative control group,the compound can significantly reduce the level of serum uric acid in rats with hyperuricemia(P<0.01);the compound had obvious inhibitory effect on immune-inflammatory factors related to gout,such as IFN-gamma,IL-1 beta,IL-2,IL-4,IL-13 and TNF-alpha.Conclusion:The compound can significantly reduce the uric acid level of hyperuricemia rat model and inhibit gout-related immune-inflammatory factors.No obvious liver and kidney damage was found in the short term.Those shows that the compound has obvious anti-inflammatory and immune effects,and has obvious therapeutic effect on hyperuricemia without toxic side effects.
