摘  要:目的:筛选并配伍出具有良好抗疲劳效果的方剂。方法:以负重游泳时间、爬杆时间为指标进行方剂筛选并优选出处方最佳配伍,为分析筛选出的最佳配伍方剂与方剂中各药材的相关性建立10批最佳配伍方剂的指纹图谱,以游泳时间、爬杆时间、游泳后的血清尿素氮(BUN)、肝糖原、血乳酸(LD)、三磷酸腺苷(ATP)、超氧化歧化酶(SOD)为指标对最佳配伍方剂进行抗疲劳活性研究与验证,通过小鼠急性毒性试验记录小鼠状况。结果:筛选出的"鹿茸酒"与"独参汤"二者的组合方能显著延长小鼠负重游泳时间以及爬杆时间,组合方的最佳配比即鹿茸∶人参∶山药=1∶3∶18缓解疲劳效果明显,能显著提高爬杆实验、负重游泳实验运动时间,提高小鼠体内肝糖原、ATP以及SOD含量,降低BUN、LD含量,建立10批最佳配比组合方提取物对照指纹图谱,从中确定9个共有峰,并全部归属到各药材,急性毒性试验表明该方剂无毒性。结论:筛选并配伍出的方剂具有良好的抗疲劳效果,且安全无毒,建立该方剂指纹图谱证明方剂与各药材间的相关性。OBJECTIVE To screen and combine the prescriptions with good anti-fatigue effect.METHODS The best formula was selected by the indexes of weight-bearing swimming time and pole climbing time.The fingerprints of 10 batches of the best formula were established to analyze the correlation between the best formula and the herbs in the formula.The antifatigue activity of the best formula was studied and verified by the indexes of swimming time,pole climbing time,serum urea nitrogen(BUN),liver glycogen,blood lactic acid(LD),adenosine triphosphate(ATP)and superoxide dismutase(SOD)after swimming.The acute toxicity test in mice was used to record the condition of mice.RESULTS The results showed that the combination of"Luzhoujiu"and"Dushen Decoction"could significantly prolong the swimming time and climbing time of mice,and the best ratio of the combination formula,i.e.,pilose antler:ginseng:yam=1:3:18,could significantly improve the exercise time of pole climbing test and swimming test,increase the contents of glycogen,ATP and SOD in the liver of mice,decrease the contents of BUN and LD,establish the control fingerprints of 10 batches of the best ratio combination formula extract,in which 9 common peaks were determined and assigned to all medicinal materials.The acute toxicity test showed that the formula had no toxicity.CONCLUSION The prescription which was screened and combined has good anti-fatigue effect and is safe and non-toxic.The fingerprint of the prescription is established to prove the correlation between the prescription and each medicinal material.
