摘  要:以肾小球系膜细胞增殖为特征的糖尿病肾病(DN)是糖尿病患者中最常见的并发症.为研究人参皂苷Rg3在DN中的作用及其体外作用机制,通过使用高糖(HG)刺激的系膜细胞增殖的体外模型,并用不同浓度梯度的人参皂苷Rg3进行干预,检测系膜细胞的增殖情况,并检测其对炎症因子MCP-1和长五聚体蛋白PTX3表达,以及NF-κB信号通路的影响.研究结果表明,人参皂苷Rg3可以一定程度地减轻HG诱导的系膜细胞增殖.在HG诱导的系膜细胞中,人参皂苷Rg3可以降低炎症因子MCP-1的表达水平.另外,人参皂苷Rg3还能显著抑制PTX3的产生,并抑制NF-κB p65的表达,增加IκBα的表达.此外,PDTC组也能显著抑制IκBα降解,降低NF-κB p65,MCP-1和PTX3的表达水平.结果表明,人参皂苷Rg3可减轻HG诱导的系膜细胞增殖,炎症反应和PTX3表达,该过程可能是通过抑制NF-κB信号通路所介导的.人参皂苷Rg3可以作为一种治疗或预防剂在DN中发挥一定的肾脏保护作用,可能与其抗炎作用和抑制PTX3的表达有关.Diabetic nephropathy(DN)characterized by proliferation of human mesangial cells is the most common complication in diabetic patients.To investigate the protective effect of ginsenoside Rg3 on DN and its mechanism in vitro,proliferation of mesangial cells is stimulated by high glucose(HG),and then intervened by different concentrations of ginsenoside Rg3.The proliferation of mesangial cells is detected,and the effects of RG3 on the expression of MCP-1,PTX3 and NF-κB signaling pathway are investigated.The results show that ginsenoside Rg3 can alleviate HG-induced mesangial cells proliferation to a certain extent.In HG-induced mesangial cells,ginsenoside Rg3 can reduce the expression of MCP-1.In addition,ginsenoside Rg3 also significantly inhibits the production of PTX3.It also inhibits the expression of NF-κB p65 and increases the expression of IκBα.In addition,the PDTC group also significantly inhibits IκBαdegradation and reduces the expression levels of NF-κB p65,IκB-α,MCP-1 and PTX3.It is concluded that,ginsenoside Rg3 can attenuate HG-induced mesangial cells proliferation,inflammatory response and PTX3 expression.This process may be mediated through inhibition of the NF-κB signaling pathway.Ginsenoside Rg3 can exert certain renal protective effects in DN,which may be related to its anti-inflammatory effect and inhibition of PTX3 expression.
