摘 要:目的:观察黄芪及发酵制剂对碘缺乏致甲状腺肿模型大鼠的氧化应激作用,以期选出药效最佳的黄芪制剂。方法:随机选择10只Wistar大鼠作为正常对照组,其余大鼠喂养碘缺乏饲料(碘含量约为20μg·kg^-1)90 d复制甲状腺肿大鼠模型,将成模大鼠随机分为模型对照组,黄芪提取液高剂量组、中剂量组、低剂量组(4.00 g·kg^-1、2.00 g·kg^-1、1.00 g·kg^-1),黄芪发酵提取液高剂量组、中剂量组、低剂量组(4.00 g·kg^-1、2.00 g·kg^-1、1.00 g·kg^-1),各组给予相应药物,连续给药36 d。于给药第30-35天使用Morris水迷宫检测大鼠空间学习记忆能力,末次给药后2 h比较各剂量组海马组织中乙酰胆碱酯酶(acetylcholinesterase,AChE)活性和氨基酸(amino acid,AA)水平,血清和海马组织中与氧化应激有关的超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、一氧化氮合酶(nitric oxide synthase,NOS)活性和一氧化氮(nitric oxide,NO)水平。结果:与正常对照组比较,模型对照组大鼠训练逃避潜伏距离明显延长(P<0.01),游泳速度、目标象限停留时间和目标象限游泳距离与游泳总距离所占比均明显减少(P<0.01);海马组织内AChE活性明显降低(P<0.01),AA水平减少(P<0.05);血清和海马组织内SOD、GSH-Px活性明显降低(P<0.01),NOS活性和NO水平明显提高(P<0.01)。与模型对照组比较,两种黄芪制剂组大鼠训练逃避潜伏距离明显缩短(P<0.01或0.05),游泳速度、目标象限停留时间和目标象限游泳距离与游泳总距离所占比均不同程度地增加(P<0.01或0.05);海马组织内AChE活性明显提高(P<0.01或0.05),AA的水平增加(P<0.05);血清和海马组织内SOD、GSH-Px活性明显升高(P<0.01或0.05),NOS活性和NO水平明显降低(P<0.01或0.05),以4.00 g·kg^-1黄芪发酵组的作用最为显著。结论:黄芪发酵制剂可明显提高碘缺乏致甲状腺肿大鼠的学习记忆能力,其作用机制�Objective:To observe the oxidative stress of jaundice and fermented preparations on thyroid-deficient model rats,in order to select the best jaundice preparation.METHODS:Ten Wistar rats were randomly selected as the normal control group.The other rats were fed with iodine-deficient diet(iodine content of about 20μg·kg^-1)for 90 days to replicate the rat model of goiter.The model rats were randomly divided into models.In the control group,high dose group,middle dose group and low dose group(4.00 g·kg^-1,2.00 g·kg^-1,1.00 g·kg^-1)of Astragalus membranaceus extract,high dose group and medium dose of Astragalus membranaceus fermentation extract Group,low-dose group(4.00 g·kg^-1,2.00 g·kg^-1,1.00 g·kg^-1),each group was given the corresponding drug,continuous administration for 36 days.The spatial learning and memory ability of rats was measured by Morris water maze on the 30 th to 35 th day after administration.The activity of acetylcholinesterase(AChE)and amino acid(AA)in hippocampus of each dose group were compared 2 h after the last administration.Levels,superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),nitric oxide(nitric oxide)associated with oxidative stress in serum and hippocampus Synthase,NOS)activity and nitric oxide(NO)levels.RESULTS:Compared with the normal control group,the training escape distance of the model control group was significantly prolonged(P<0.01),and the swimming speed,target quadrant stay time and target quadrant swimming distance and swimming total distance were significantly reduced(P<0.01);AChE activity in hippocampus was significantly decreased(P<0.01),AA level was decreased(P<0.05);SOD and GSH-Px activities in serum and hippocampus were significantly decreased(P<0.01),NOS activity and NO level were significantly decreased;NOS activity and NO levels are significantly improved(P<0.01).Compared with the model control group,the training escape distance of the two groups of the Huangqi preparation group was significantly shortened(P<0.01 or 0.05),and the swimming speed,the t
