摘  要:目的探讨木犀草素对哮喘幼鼠的防治作用及可能机制。方法36只BALB/c幼鼠,雌雄不限,随机分为对照组、哮喘组和木犀草素组,卵蛋白致敏的方法制备哮喘幼鼠模型。最后一次激发后24h,处死大鼠后取肺组织,HE染色观察各组幼鼠肺组织病理学改变;取大鼠血清检测SOD、MDA、GSH-Px及T-AOC变化;采用实时PCR和Western-blot法检测各组幼鼠肺组织COX-2、PGE2、TNF-α及IL-1βmRNA和蛋白表达。结果与对照组相比,哮喘组幼鼠气道上皮组织结构紊乱,肺内可见大量炎性细胞浸润,木犀草素明显减轻哮喘幼鼠肺内炎性浸润,降低哮喘幼鼠血清MDA含量,上调SOD、GSH-Px、T-AOC水平,下调哮喘幼鼠肺组织COX-2、PGE2、TNF-α及IL-1β的mRNA和蛋白表达(P<0.01)。结论木犀草素减轻哮喘幼鼠气道炎症,与其抗氧化应激和抑制COX-2信号通路相关。Objective To investigate the therapeutic effect and possible mechanism of luteolin on the asthmatic young mice.Methods 36 BALB/c mice,male and female,were randomly divided into control group,asthma group(established by ovalbumin)and luteolin group.24 hours after the last stimulation,the lung tissues of the mice were sacrificed,and the pathological changes of lung tissues were observed by HE staining.The contents of SOD,MDA,GSH-Px and T-AOC in serum were detected.The expressions of COX-2,PGE2,TNF-αand IL-1βwere determined by real-time PCR and Western-blot.Results Compared with the control group,a large number of inflammatory infiltration cells and disordered epithelium in the lung tissues were found in the asthmatic group.Luteolin alleviated the inflammatory infiltration,down-regulated MDA content in serum and the expression levels of COX-2,PGE2,TNF-αand IL-1βin lung tissue,upregulated the levels of SOD,GSH-Px and T-AOC in lung tissue of asthmatic young mice(P<0.01).Conclusion Luteolin inhibited the airway inflammation in asthmatic young mice,which is related to antioxidant stress and inhibition of COX-2 signaling pathway.
