摘 要:目的评价沉默MTA1基因对神经胶质瘤细胞U-87-MG侵袭与迁移能力的影响。方法应用慢病毒转染技术沉默MTA1基因的表达,通过Western blot实验与荧光共聚焦实验检测MTA1蛋白与F-actin蛋白的表达;通过Transwell实验评估细胞侵袭与迁移能力;Western blot实验检测E-cadherin,N-cadherin蛋白的表达。结果沉默MTA1基因可下调F-actin蛋白的表达水平,抑制U-87-MG细胞的侵袭与迁移能力,并且上调N-cadherin蛋白,下调E-cadherin蛋白的表达。结论沉默MTA1基因抑制胶质母细胞瘤细胞系U-87-MG的侵袭与迁移能力与影响F-actin的集聚与EMT相关蛋白N-cadherin与E-cadherin表达相关。Objective To evaluate the effect of silencing MTA1 on the invasion and migration of glioblastoma cell line U-87-MG. Methods The expression of MTA1 was silenced by lentiviral transfection technique. The expression of MTA1 protein and F-actin protein was detected by Western blot and fluorescence confocal assay. The invasion and migration abilities of U-87-MG cells were evaluated by Transwell assay. The expression of E-cadherin and N-cadherin protein was detected by Western blot. Results Silencing MTA1 down-regulated the expression level of F-actin protein, inhibited the invasion and migration of U-87-MG cells, and up-regulated the expression levels of N-cadherin protein, and downregulated the expression levels of E-cadherin protein. Conclusion Silencing MTA1 inhibits the invasion and migration of glioblastoma cell line U-87-MG by affecting the accumulation of F-actin and the expression of EMT-related proteins Ncadherin and E-cadherin.
