乌头注射液对KOA模型兔软骨细胞ADAMTS-4 ADAMTS-5表达影响的实验研究

摘　　要：目的:观察乌头注射液对膝骨性关节炎模型兔膝关节软骨细胞中ADAMTS-4、ADAMTS-5表达的影响。方法:采用随机分组的方式,将40只新西兰大耳白兔分为正常、模型(生理盐水)、阳性对照(玻璃酸钠)、乌头注射液组4组,除正常组外其他3组兔膝关节进行关节炎造模。造模完成后,分别于兔膝关节腔内按照0.1 mL/kg的剂量标准注射生理盐水、玻璃酸钠、乌头注射液。饲养1周后处死取材,采用免疫组化法观察软骨细胞中ADAMTS-4、ADAMTS-5表达变化情况。结果:模型组兔膝关节软骨细胞中ADAMTS-4、ADAMTS-5表达较正常组有显著升高;乌头注射液组软骨细胞中ADAMTS-4、ADAMTS-5的表达较模型组组显著降低(P<0.05),与阳性组比较差异明显(P<0.05)。结论:乌头注射液能够抑制膝骨性关节炎兔膝关节软骨细胞中ADAMTS-4、ADAMTS-5的表达,保护膝关节软骨。Objective:To observe the effects of Aconite Injection on adamts-4 and adamts-5 expressions in articular chondrocytes of rabbits with knee models of knee osteoarthritis.Methods:40 New Zealand white rabbits were divided into four groups randomly:normal,model,positive control and Aconite Injection groups.After the modeling was completed,saline,Sodium hyaluronate Injection and Aconitum Injection were injected into the rabbits’knee cavity of the model,positive control and Aconite Injection group at the standard dose of 0.1 mL/kg respectively.After feeding for a week,the expression changes of adamts-4 and adamts-5 in chondrocytes were observed by immunohistochemistry.Results:In the model group,the expressions of adamts-4 and adamts-5 in articular chondrocytes were significantly increased;in the Aconitum Injection group,the expressions of adamts-4 and adamts-5 in articular chondrocytes were significantly decreased,the differences between Aconitum Injection group and positive group were significance(P<0.05).Conclusion:Aconite Injection can suppress the expressions of adamts-4 and adamts-5 in articular chondrocytes of rabbits with KOA,so that it can protect the knee cartilage of rabbits.
