摘 要:目的探讨细粒棘球蚴P29重组抗原免疫小鼠后其脾脏和外周血中免疫细胞数量的动态变化,为分子疫苗的转化应用提供参考资料和理论依据。方法30只BALB/c小鼠分为空白对照组、佐剂组和P29组,佐剂组注射等量佐剂,P29组小鼠腹部皮下注射P2910μg+佐剂,进行两次免疫(第0、2周);采用流式细胞术动态检测小鼠脾脏和外周血免疫细胞的比例变化。结果(1)脾脏中P29组第1、2、3、4、6周CD8+T细胞数均高于对照组和佐剂组(P均<0.05)。P29组第3周CD4+T/CD8+T比值高于对照组(P<0.05),第1、2、3、4、6周低于佐剂组(P<0.05)。外周血中P29组第1、2、3、4、6周CD8+T细胞数均高于对照组(P均<0.05),第3、6周高于佐剂组(P均<0.05)。P29组第3、6周和佐剂组第1、2、4周CD4+T/CD8+T比值均低于对照组,第1、2、3、4、6周低于佐剂组(P均<0.05)。(2)脾脏中P29组第1、2、3周MDSCs占单个核细胞数量百分比均高于对照组和佐剂组(P均<0.05)。外周血中P29组第1、4周MDSCs占单个核细胞数量百分比均高于对照组(P<0.05或P<0.01),第1周高于佐剂组(P<0.01)。(3)脾脏中P29组第2周NK细胞数量占单个核细胞数量的百分比均高于对照组和佐剂组(P均<0.05)。外周血中P29组小鼠第1、2、4周NK细胞数量占单个核细胞数量的百分比均高于对照组(P均<0.05),第1、2、3、4周均高于佐剂组(P均<0.05)。结论细粒棘球蚴重组抗原P29疫苗诱导小鼠免疫保护过程中,脾脏和外周血的CD8+T、NK及MDSCs细胞数量上调。Objective To investigate the dynamic changes of immune cells induced by Echinococcus granulosus(E.g)recombinant P29 antigen,and provide reference and theoretical basis for the transformation of molecular vaccines.Methods 30 BALB/c mice were subcutaneously injected with 10μg recombinant vaccine P29 twice(0 week and second week)to induce immune cell differentiation.Flow cytometry was used to dynamically detect the proportion variety of immune cells in control,adjuvant and P29 groups.Results After immunization of mice with P29 recombinant antigen,spleen index and number of mononuclear cells of the mice from the 2 nd week to the 6 th week was significantly increased in the P29 group compared with the adjuvant group and the control group.The number of cell CD4+T decreased and that of CD8+T cell increased significantly from the 1 st week to the 6 th week,The ratio of CD4+T/CD8+T decreased.The number of myeloid-derived suppressor cells(MDSCs)in the spleen increased significantly from the 1 st week to the 4 th week,and the number of MDSCs in the peripheral blood increased significantly from the 1 st week to the 3 rd week.The number of NK cells in the peripheral blood increased significantly from the 1 st week to the 6 th week.Conclusion T cells,NK cells and MDSCs are significantly up-regulated after E.g recombinant antigen P29 vaccination.
