摘 要:以脱辅基肌红蛋白质和还原性溶菌酶为研究对象,考察了射频透镜1电压等质谱条件对蛋白质ESI-MS图谱荷电状态分布(CSD)的影响。研究发现:在“较软”ESI条件下射频透镜1电压能在一定程度上显著改变ESI-MS图谱右左两侧两个分布的相对强度,而右左两侧两个电荷中心的相对位置基本保持不变,这种影响与溶液相中蛋白质分子的构象类型密切有关,本文对这一现象的机理做了探讨。Apomyoglobin and reduced lysozyme were chosen as the research objects,and the effects of MS instrumental parameters such as RF Lens 1 voltage on charge state distribution(CSD)of ESI-MS spectrum were investigated.It was found that under the condition of"soft"ESI instrumental settings,the voltage of RF lens 1 could significantly change the relative intensity of the two distributions on the right and left sides of ESI-MS spectrum,while the relative positions of the two charge centers on the right and left sides remained basically unchanged.The effect of RF Lens 1 voltage was closely related to the conformation of proteins in the bulk solution,and the mechanism was discussed.
