摘 要:目的:研究探讨全真一气汤多糖对小鼠缺氧模型的氧化应激干预作用。方法:小鼠随机分为正常对照组、全真一气汤浸膏高剂量组、全真一气汤浸膏低剂量组、全真一气汤多糖高剂量组、全真一气汤多糖低剂量组,每组小鼠连续灌胃8 d,末次灌胃2 h后,采用常压密闭缺氧实验、亚硝酸钠中毒实验、急性脑缺血性缺氧实验,记录小鼠存活时间。对于急性脑缺血性缺氧受试小鼠,检测氧化应激指标如总抗氧化能力(T-AOC)、总超氧化歧化酶(T-SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽(GSH)、一氧化氮合酶(TNOS、iNOS)活力、羟脯氨酸(Hyp)含量等,并结合肺脏的病理切片进行抗氧化作用研究。结果:全真一气汤组及多糖组小鼠耐缺氧能力和耐急性脑缺血性缺氧能力均比正常对照组强。全真一气汤的多糖对于T-AOC、GSH、TNOS活力和iNOS活力均有显著作用,浸膏对T-AOC和Hyp有显著作用(P<0.05或P<0.01)。结论:全真一气汤组及多糖组小鼠在常压密闭缺氧实验、亚硝酸钠中毒实验、急性脑缺血性缺氧实验的存活时间均比正常对照组强及对肺部的显著保护作用,表明全真一气汤及多糖具有一定的抗氧化应激干预作用。Objective:To investigate the anti-oxidation effect of polysaccharide(PS)of Quanzhen Yiqi Formula on oxidative stress in hypoxia mice.Method:Three pharmacological experiments were performed with hypoxia mice,hypoxic tolerance in a sealed environment under norm pressure,acute poisoning by sodium nitrite and hypoxic tolerance with acute ischemic brain.The survival time of each mouse was determined and the biochemical indicators such as T-AOC,T-SOD,MDA,CAT,GSH,NO,TNOS activity,iNOS activity and Hyp were detected as well as pathological sections.Results:Quanzhen Yiqi Formula and PS could significantly improve the hypoxic tolerance of mice in a sealed environment under norm pressure and that with acute ischemic brain.It showed high anti-oxidation activity on T-AOC,GSH,TNOS activity and iNOS activity.The pathological sections could also show the anti-oxidation effect of the extract and PS.Conclusion:The PS of Quanzhen Yiqi Formula has significant activity against oxidative stress in hypoxia mice.
