摘  要:目的探讨黄芪灌胃给药C57BL/6小鼠后脾脏中淋巴细胞及其相关表面分子的变化。方法将黄芪通过灌胃给药小鼠,6d后将对照组和黄芪组小鼠的脾细胞进行分离,制备单细胞悬液,然后通过流式细胞术检测小鼠B细胞、T细胞、CD8+T细胞和NK细胞及其相关表面分子CXCR3、CD69和CD62L的百分比含量。结果黄芪组小鼠脾脏中B细胞和NK细胞的百分比显著升高(P<0.05),而T细胞和CD8+T细胞的百分比显著降低(P<0.05和P<0.01)。黄芪组CXCR3分子在NK细胞中的表达明显高于对照组(P<0.05);黄芪组CD69分子在CD8+T细胞和NK细胞中的表达均明显高于对照组(P<0.05);黄芪组CD62L分子在B细胞、T细胞、CD8+T细胞和NK细胞中的表达均显著低于对照组(均P<0.01)。结论C57BL/6小鼠在给药黄芪后,其脾脏中上述淋巴细胞及其相关表面分子可发生不同程度的变化,提示黄芪对小鼠的免疫功能可以产生多种调节作用。Astragali radix,a medicinal plant,has the functions of reinforcing qi and strengthening spleen.Here,we designed this study to explore the effect of Astragali radix on the immune function of C57BL/6 mice.Astragali radix was given to mice by intragastric administration for 6 days,and then,the spleen cells of the normal control mice and Astragali radix treated mice were isolated to make single cell suspensions.Fluorescence activated cell sorting(FACS)was used to detect the percentage of B cells,T cells,CD8+T cells.NK cells and their related surface molecules CXCR3,CD69 and CD62L.Data showed that the percentage of B cells and NK cells in the spleen of the Astragali radix treated mice were increased significantly(P<0.05),while the percentage of T cells and CD8+T cells were decreased significantly(P<0.05 and P<0.01),as compared with control group.Furthermore,the expression of CXCR3 in NK cells of Astragali radix group was significantly higher than that in the control group(P<0.05);the level of CD69 expressed by CD8+T cells and NK cells were increased significantly(P<0.05);and the expression of CD62L in B cells,T cells,CD8+T cells and NK cells were decreased significantly(P<0.01,P<0.05,P<0.05 and P<0.05).Taken together,intragastric administration of Astragali radix results in different changes of the lymphocytes and their related surface molecules in the spleen of C57BL/6 mice,suggesting that Astragali radix can produce various adjustments on the immune function of mice.
