摘 要:[目的]探讨甘蔗叶多糖对血瘀证下急性心肌梗死模型大鼠的治疗作用及其机制.[方法]取Wistar大鼠随机分为正常对照组、模型对照组、阳性对照组(阿司匹林肠溶片5mg·kg^-1)和甘蔗叶多糖大、中、小剂量(20.0,10.0,5.0mg·kg^-1)组,给予相应处理.采用皮下注射大剂量盐酸肾上腺素复加冰水冷浴方法建立大鼠急性血瘀模型,在急性血瘀情况下结扎左冠状动脉前降支复制急性心肌梗死模型,检测各组大鼠血液流变学指标、心肌酶水平及心肌超微结构的变化.[结果]与模型对照组比较,甘蔗叶多糖各剂量组体外血栓长度明显缩短,体外血栓湿质量和干质量显著降低,血液黏度降低,红细胞压积及血小板聚集率明显受到抑制,凝血时间延长,天冬酸氨基转移酶、肌酸激酶及乳酸脱氢酶水平降低,相比较差异均有统计学意义(P<0.05,P<0.01,P<0.001),心肌细胞超微结构得到改善.[结论]甘蔗叶多糖对大鼠急性血瘀证下心肌梗死具有保护作用,其可能机制认为与它增强抗氧化酶活性、抑制体外二磷酸腺苷诱导的血小板聚集及改善细胞膜结构有关系.OBJECTIVE To explore the therapeutic effects of sugarcane leaf polysaccharide on acute myocardial infarction model rats with blood stasis syndrome and its mechanism.METHODS Wistar rats were randomly divided into normal control group,model control group,positive control group(aspirin enteric-coated tablet,5mg·kg^-1)and high,middle,low sugarcane leaf polysaccharide group(20.0,10.0 and 5.0mg·kg^-1).The acute blood stasis model of rats was established by subcutaneous injection of large dose of adrenaline hydrochloride combined with ice water cold bath.The acute myocardial infarction model was established by ligating the anterior descending branch of left coronary artery under the condition of acute blood stasis.The changes of hemorheology,myocardial enzyme level and myocardial ultrastructure in each group were detected.RESULTS As compared with the model control group,the length of thrombus in vitro in sugarcane leaf polysaccharide groups was significantly shortened,the wet and dry mass of thrombus in vitro were obviously reduced,blood viscosity decreased,hematocrit and platelet aggregation rate were obviously inhibited,coagulation time increased,the levels of aspartate aminotransferase,creatine kinase and lactate dehydrogenase decreased,the differences were statistically significant(P<0.05,P<0.01,P<0.001),and myocardial cell ultrastructure was improved.CONCLUSIONSugarcane leaf polysaccharide had protective effects on myocardial infarction in rats with acute blood stasis syndrome,and its possible mechanism was thought to be related to its enhancement of antioxidant enzyme activity,inhibition of platelet aggregation induced by adenosine diphosphate in vitro and improvement of cell membrane structure.
