咳喘凝胶贴膏的初步药效学研究

摘　　要：目的:观察皮肤外用制剂咳喘凝胶贴膏的镇咳、平喘和化痰的作用,为传统黑膏药制剂的剂型改进和工艺优化提供药理学依据。方法:昆明种小鼠40只,随机分成高剂量样品组(含生药量18 g·kg^-1)、低剂量样品组(含生药量9 g·kg^-1)、空白对照组、阳性对照组,每组10只。采用氨水诱发小鼠咳嗽法和小鼠气管段酚红排泌法,考察咳喘凝胶贴膏的镇咳、平喘作用;豚鼠40只,同上分组方法,采用乙酸胆碱(ACH)和磷酸组胺(HA)诱发豚鼠哮喘试验,考察咳喘凝胶贴膏的平喘作用。结果:咳喘凝胶贴膏高、低剂量组均可明显抑制小鼠的咳嗽次数,且高剂量组抑制作用较阳性对照组更明显(P<0.05);用药后4 h,高剂量组可明显延长引喘潜伏期(P<0.05);高、低剂量及阳性组均能够明显增加小鼠呼吸道酚红的排泌量,且高、低剂量组更优于阳性对照组(P<0.05)。结论:咳喘凝胶贴膏具有一定的镇咳、平喘和化痰作用,为中医院传统黑膏药敷贴的剂型更迭提供实验依据。Objective:To observe the antitussive,antiasthmatic and phlegm-resolving effects of Kechuan gel plasters,a traditional Chinese medicine preparation for skin topical application,in order to provide pharmacological basis for the improvement of dosage forms and process optimization of traditional black plaster preparations in our hospital.Methods:Three animal models were established.The cough model was induced by ammonia in mice,the asthma model was induced by ACH and histamine phosphate(HA)in guinea pigs,and the phenol red excretion in the trachea of mice was also observed to respectively investigate the antitussive,antiasthmatic and expectorant effects of Kechuan gel plasters.Results:Kechuan gel plasters could prolong the incubation period of cough induced by concentrated ammonia in mice,extend the incubation period of induced asthma response induced by ACH and HA in guinea pigs,and increase the secretion of phenol red in mouse gas duct.Conclusion:Kechuan gel plasters have certain therapeutic effect on cough,asthma and phlegm,which provide experimental basis for the improvement of traditional black plasters in traditional Chinese medicine hospitals.
