摘 要:目的:明确水通道蛋白1(AQP1)基因在复张性肺水肿(RPE)中的表达,探讨AQP1在RPE发病过程中的作用及机制。方法:2016年9月至2018年9月,选取AQP1野生型小鼠和AQP1基因敲除型小鼠为研究对象,按Sakao改良方法建立鼠RPE模型,在复张前及复张后30 min收集支气管肺泡灌洗液及血样,测定总蛋白、二棕榈酰磷脂酰胆碱(DPPC)、白细胞介素–8(IL–8)、肿瘤坏死因子(TNF–α)含量。结果:AQP1蛋白电泳灰度分析,各组内AQP1–与AQP1+标本相比较,差异具有统计学意义(P <0.05),而模型组与乌司他丁联合模型组AQP1+标本结果比较,差异无统计学意义(P> 0.05)。与AQP1野生型小鼠相比,AQP1基因敲除小鼠的AQP1蛋白表达水平显著降低,与假手术组比较,DPPC、IL–8和TNF–α在模型组与乌司他丁模型组处于显著高表达水平,乌司他丁模型组又低于模型组,差异均具有统计学意义(P <0.05),但乌司他丁并不能影响AQP1的表达,差异无统计学意义(P> 0.05)。结论:AQP1的表达与RPE的程度有关,乌司他丁能减轻RPE的程度,但与AQP1的表达无关。Objective To clarify the expression of aquaporin 1(AQP1) gene in reexpansion pulmonary edema(RPE), and to explore the role and mechanism of AQP1 in the pathogenesis of reexpansion pulmonary edema. Method From September 2016 to September 2018, AQP1 wild-type mice and AQP1 gene knockout mice were selected as the research objects. The model of reexpansive pulmonary edema was established by modified Sakao method. Bronchoalveolar lavage fluid and blood samples were collected before and 30 minutes after reexpansion. Totalprotein, dipalmitoyl phosphatidylcholine(DPPC), interleukin–8(IL–8) and tumor necrosis factor–A(TNF–α) were determined. Result The gray level analysis of AQP1 protein electrophoresis showed that there were significant differences between AQP1-and AQP1 + samples in each group(Ρ < 0.05), while the results of AQP1 + samples in model group and ulinastatin combined model group were compared(Ρ > 0.05). Compared with wild-type AQP1 mice, the expression of AQP1 protein in AQP1 knockout mice was significantly lower(Ρ < 0.05). Compared with sham-operated mice, DPPC, IL–8 and TNF–αwere significantly higher in model group and ulinastatin model group(Ρ < 0.05). The ulinastatin model group was lower than the model group(Ρ < 0.05), but it did not affect the expression of AQP1(Ρ > 0.05). Conclusion This study showed that the expression of AQP1 was related to the degree of reexpansive pulmonary edema. Ulinastatin could alleviate the degree of reexpansive pulmonary edema, but it has no obvious relationship with the expression of AQP1.
