摘 要:目的:通过观察有氧运动或抗阻运动干预对大鼠脂肪和下丘脑组织中骨形态发生蛋白7(BMP7)表达的影响,结合大鼠在静息状态下能量消耗的变化,探讨不同运动方式通过BMP7调控大鼠能量代谢的作用。方法:将30只8周龄雄性SD大鼠随机分为三组:安静对照组(CON组)、有氧运动组(AE组)、抗阻运动组(RE组),每组各10只。AE组大鼠利用跑台进行8周的有氧运动训练,每周训练5天,跑台速度和训练时间从第1周的15 m/min、20 min递增至第8周的28 m/min、60 min。RE组大鼠利用"负重爬梯法"进行8周的抗阻运动训练,每2天训练一轮,抗阻负荷根据训练情况递增。完成最后一次训练24小时后,利用动物气体代谢分析仪测试大鼠的静息能量消耗水平。然后,检测大鼠血清BMP7水平,检测腹股沟皮下白色脂肪(WAT)、肩胛间棕色脂肪(BAT)及下丘脑BMP7基因和蛋白表达水平,检测BAT中UCP1基因和蛋白表达水平。结果:运动干预结束后,CON组大鼠体重显著高于AE组和RE组(P<0.01),CON组的血清BMP7水平显著低于AE组(P<0.05)。在WAT中,CON组BMP7 m RNA表达量显著低于AE组(P<0.01),且其蛋白表达量显著低于AE组和RE组(P<0.05)。在BAT中,CON组BMP7及UCP1的mRNA表达量均显著低于AE组和RE组(P<0.05,P<0.01),但蛋白表达量无显著差异。在下丘脑中,CON组BMP7的mRNA及蛋白表达量均显著低于AE组(P<0.05)。此外,CON组大鼠在静息状态下的氧耗量及能量消耗均显著低于AE组和RE组(P<0.05,P<0.01)。结论:8周有氧运动或抗阻运动干预均提高了大鼠静息能量消耗水平并减缓了体重增长幅度,这可能与运动诱导脂肪组织高表达BMP7进而促进了白色脂肪棕色化并提高了BAT产热水平有关。相对于抗阻运动,有氧运动提高了大鼠下丘脑组织中BMP7的表达水平,可能通过下丘脑影响了大鼠的摄食行为,但还需更多研究阐释BMP7在调节能量摄入中的作用及机制。Objective To investigate the effect of aerobic and resistance exercise training on resting energy expenditure by observingthe expression of the bone morphogenetic protein(BMP7)in rats.Methods Thirty male Sprague-Dawley rats were randomly divided into a control group(CON,n=10),an aerobic exercise group(AE,n=10)and a resistance exercise group(RE,n=10).The AE group completed an 8-week aerobic exercise training program by running on a treadmill for 5 days/week,with the running speed and duration of 15 m/min and 20 minutes at the 1 stweek increasing to 28 m/min and 20 minutes at the 8 thweek.The RE group climbed a ladderevery other day for 8 weeks,and the load was adjusted according to the rats’capacity in each training session.Twenty-four hours after the last training,the resting energy expenditure of all rats was measured using the comprehensive lab animal monitoring system(made by Columbus Instruments,Columbus,USA).Then the serum was collected to measure the mRNA and protein levels of BMP7 and UCP1 of the white adipose tissue(WAT),brown adipose tissue(BAT)and hypothalamus.Results After the training program,the body weight of CON group was significantly higher than both AE and RE groups(P<0.01),while the serum BMP7 level of the former group was significantly lower than the AE group(P<0.05).As observed in WAT,the BMP7 mRNA level of the CON group was significantly lower compared with AE group(P<0.01),and its protein level was significantly lower than both AE and RE groups(P<0.05).In BAT,additionally,the CON group was significantly lower in the mRNA levels of BMP7 and UCP1 compared with AE and RE groups(P<0.05,P<0.01),but no significant difference was observed in the protein level.In the hypothalamus,both the BMP7 m RNA and protein level of CON group were significantly lower than AE group(P<0.05).Moreover,CON group had significantly lower resting oxygen consumption and energy expenditure than both AE and RE groups(P<0.05,P<0.01).Conclusion The 8-week aerobic or resistance exercise training increases the resting energ
