摘 要:【目的】探讨银杏叶分散片(ginkgo biloba leaf dispersible tablet,GBT)对局灶性脑梗死大鼠的保护作用及可能的作用机制。【方法】大鼠随机分为假手术组、模型组、阳性对照组(尼莫地平)、低剂量银杏叶分散片组(L-GBT组)、中剂量银杏叶分散片组(M-GBT组)和高剂量银杏叶分散片组(H-GBT组)。采用改良Longa法建立局灶性脑梗死大鼠模型。神经功能缺损评分观察大鼠行为学,TTC法染色并计算脑梗死体积,TUNEL染色观察细胞凋亡情况,酶联免疫吸附法检测脑组织炎性因子肿瘤坏死因子(tumor necrosis factor,TNF-α)、IL-1β和IL-6表达水平,Western blotting检测Bax、Bcl-2、p-ERK1/2和p-NF-κB蛋白表达。【结果】与假手术组比,模型组大鼠神经功能缺损评分、脑梗死体积、脑组织细胞凋亡指数、TNF-α、IL-1β和IL-6水平以及Bax、p-ERK1/2和p-NF-κBp65蛋白表达均显著增加(P<0.05),Bcl-2蛋白表达显著减少(P<0.05)。与模型组比,M-GBT组、H-GBT组和阳性对照组大鼠神经功能缺损评分、脑梗死体积、脑组织细胞凋亡指数、TNF-α、IL-1β和IL-6水平以及Bax、pERK1/2和p-NF-κBp65蛋白表达均显著减少(P<0.05),Bcl-2蛋白表达显著增加(P<0.05);L-GBT组各检测指标差异不显著(P>0.05)。H-GBT组和阳性对照组各检测指标比较差异也不显著(P>0.05)。【结论】GBT可能通过抑制ERK1/2/NF-κB信号通路的激活,下调TNF-α、IL-1β、IL-6等炎性因子的表达,减轻局灶性脑梗死大鼠脑组织损伤。【Objective】To explore the protective effect of Ginkgo biloba leaf dispersible tablet(GBT)on rats with focal cerebral infarction and its possible mechanism.【Methods】Rats were randomly divided into sham operation group,model group,positive control group(nimodipine),low-dose Ginkgo biloba leaf dispersible tablet group(L-GBT group),medium-dose Ginkgo biloba leaf dispersible tablet group(M-GBT group)and high-dose Ginkgo biloba leaf dispersible tablet group(H-GBT group).The rat model of focal cerebral infarction was established by modified Longa method.The neurological deficit score was used to observe the behaviors of rats.TTC staining was used to calculate the volume of cerebral infarction and apoptosis was observed by TUNEL staining.The expression levels of inflammatory factors TNF-α,IL-1βand IL-6 in brain tissues were detected by ELISA.Western blotting was used to detect the protein expression of Bax,Bcl-2,p-ERK1/2 and p-NF-κB.【Results】Compared with the sham operation group,the neurological deficit score,cerebral infarction volume,apoptotic index of brain tissues,levels of TNF-α,IL-1βand IL-6,and the protein expression of Bax,p-ERK1/2 and p-NF-κBp65 in the model group were significantly increased(P<0.05),while the protein expression of Bcl-2 was significantly decreased(P<0.05).Compared with the model group,the neurological deficit score,cerebral infarction volume,apoptotic index of brain tissues,levels of TNF-α,IL-1βand IL-6,and the protein expression of Bax,p-ERK1/2 and p-NF-κBp65 in the M-GBT group,H-GBT group and positive control group decreased significantly(P<0.05),while the protein expression of Bcl-2 increased significantly(P<0.05).There was no significant difference in the index detection of L-GBT group(P>0.05).There was no significant difference between H-GBT group and positive control group(P>0.05).【Conclusion】GBT may reduce brain tissue injury in rats with focal cerebral infarction by inhibiting the activation of ERK1/2/NF-κB signaling pathway and down-regulating the expression
