滋阴清热药和强的松对狼疮小鼠血清ABTS、MDA及抗ds-DNA抗体的影响

摘　　要：目的研究滋阴清热药和强的松对MRL/lpr狼疮小鼠血清2,2-联氮-双(3-乙基苯并噻唑-6-磺酸)(ABTS)、丙二醛(MDA)及抗双链DNA(ds-DNA)抗体的影响。方法将24只雌性12周龄的MRL/lpr狼疮小鼠随机分为4组:滋阴清热药组、强的松组、滋阴清热药和强的松合用组、空白对照组。采用滋阴清热药按0.02 mL·g-1剂量对滋阴清热药组灌胃,每日1次;采用0.3 g·L-1强的松药液按0.02 mL·g-1剂量对强的松组灌胃,每日1次;采用滋阴清热药和强的松药液对合用组灌胃,方法和剂量同前;采用0.6 mL生理盐水对空白对照组灌胃,每日1次。观察3个月,抽取小鼠血清检测ABTS、MDA和抗ds-DNA抗体水平。结果滋阴清热药组ABTS水平高于其他3组,差异有统计学意义(均P<0.05);强的松组ABTS水平低于空白对照组,差异有统计学意义(P<0.05)。强的松组MDA水平高于其他3组,差异有统计学意义(均P<0.05)。滋阴清热药组、强的松组、合用组抗ds-DNA抗体水平均低于空白对照组,差异有统计学意义(均P<0.05);滋阴清热药组、强的松组、合用组抗ds-DNA抗体水平比较,差异无统计学意义(均P>0.05)。结论滋阴清热药能显著增强抗氧化能力,强的松能显著降低抗氧化能力,两者均能引起抗ds-DNA抗体水平下降,提示滋阴清热药治疗系统性红斑狼疮(SLE)有不同于糖皮质激素的作用机制,体现了中医药治疗SLE的特色。Objective To research the effects of nourishing yin and clearing heat medicine and prednisone on serum 2,2-Azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS),malondialdehyde(MDA),anti-double stranded DNA antibody(anti-ds-DNA antibody)of MRL/lpr lupus mice.Methods A total of 24 female 12-week-old MRL/lpr lupus mice were randomly divided into four groups:nourishing yin and clearing heat medicine group,prednisone group,nourishing yin and clearing heat medicine group combined prednisone group,blank control group.Nourishing yin and clearing heat medicine group was given nourishing yin and clearing heat drugs at 0.02 mL·g-1 by gastric perfusion,once per day.Prednisone group was given 0.3 g·L-1 prednisone solution at 0.02 mL·g-1 by gastric perfusion,once per day.Combination group was given nourishing yin and clearing heat medicine and prednisone solution,and the method and dose were the same as before.Blank control group was given 0.6 mL saline by gastric perfusion,once per day.After 3 months of observation,the serum of mice was extracted to detect the levels of ABTS,MDA and anti-ds-DNA antibody.Results The ABTS of nourishing yin and clearing heat medicine group was higher than those of the other three groups,and the differences were statistically significant(all P<0.05).The ABTS of prednisone group was lower than that of blank control group,and the difference was statistically significant(P<0.05).The MDA of prednisone group was higher than those of the other three groups,and the differences were statistically significant(all P<0.05).The levels of serum anti-ds-DNA antibody of nourishing yin and clearing heat medicine group,prednisone group,combination group were all lower than that of blank control group,and the differences were statistically significant(all P<0.05).There were no significant differences in anti-ds-DNA antibody levels among nourishing yin and clearing heat medicine group,prednisone group and combination group(all P>0.05).Conclusion Nourishing yin and clearing heat medicine can significantly enhance
