摘  要:目的检测大鼠睾丸发育中SCN1B基因的转录表达及定位情况,探讨在精子发生中该基因可能的作用机制。方法选取出生后第2~65天总共21个时间点的Wistar雄性大鼠,每个时间点取三只来自不同笼鼠的睾丸组织,提取总的mRNA并逆转录成cDNA,采用实时定量PCR技术检测SCN1B基因在大鼠睾丸组织中的转录表达。另选取60日龄鼠的睾丸组织,采用免疫组织化学方法观察SCN1B基因的蛋白表达定位情况。结果鼠出生第8天后SCN1B基因转录表达开始逐渐上升,在第16天达到最高值,随后到65天缓慢降低,但表达仍然较高。60日龄睾丸曲细精管中的精子细胞有阳性表达。结论大鼠睾丸组织发育的中后期,SCN1B基因的转录表达高,可能在减数分裂和精子成熟时期起着一定作用,且该基因在精子细胞中有表达。Objective To detect the transcriptional expression and localization of SCN1B gene in rat testis development,and to explore the possible mechanism of this gene in spermatogenesis.Methods Male Wistar rats at 21 time points from day 2-65 after birth were selected.Three rats from different cages were taken at each time point to obtain testicular tissues.Total mRNA was extracted and reverse transcribed into cDNA.Quantitative PCR was used to detect the transcriptional expression of SCN1B gene in rat testis tissue.The position of protein edit by SCN1B gene was observed by immunohistochemistry in 60 days old testis tissue.Results The transcriptional expression of SCN1B gene began to increase gradually after the 8th day of birth,reached the highest value at the 16th day,and then slowly decreases since the 65th day,but the expression was still at a high level.The SCN1B gene was positively expressed in sperm cells in the seminiferous tubules of testicular curvature of 60-day-old rats.Conclusions In the middle and late stages of rat testis development,the transcriptional expression of SCN1B gene is at a high level,which may play a role in meiosis and sperm maturation,and the gene is expressed in sperm cells.
