壁虎/蝉拟青霉发酵菌质粗提物抗肿瘤研究

摘　　要：目的:探索中药壁虎经蝉拟青霉发酵后菌质粗提物中蛋白质及体外抗肿瘤活性的变化。方法:分别用冻融、超声、Tris-HCl提取壁虎和壁虎/蝉拟青霉菌质抗肿瘤活性成分,通过蛋白质浓度检测、十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析壁虎发酵前后蛋白含量与分子量的变化;MTT检测壁虎发酵前后抗HepG2肿瘤细胞活性的变化及不同提取方式抗肿瘤活性的差异。结果:壁虎发酵后菌质3种提取方式所得蛋白质含量均减少约30%,分子量>10 kDa的蛋白电泳条带均消失;体外抗HepG2肿瘤细胞增殖活性增加,且超声提取物诱导HepG2肿瘤细胞凋亡的IC 50值为10.769 mg·mL-1,比发酵前降低了约50%。结论:蝉拟青霉的生长代谢分解了壁虎中的部分蛋白,且壁虎经蝉拟青霉发酵后抗HepG2细胞增殖活性增强。Objective:To explore the changes of protein and antitumor activity in crude extract of Chinese medicine Gekko swinhonis fermented by Cordyceps Cicadae in vitro.Methods:The proteins in G.swinhonis and G.swinhonis/Paecilomyces cicadae fermentation substance were extracted by freeze-thaw,ultrasonic and Tris-HCl methods.Analysis of protein content and molecular weight of G.swinhonis before and after fermentation by protein concentration detection and SDS-PAGE.MTT assay for detection of anti-HepG2 tumor cell activity before and after G.swinhonis fermentation and differences in antitumor activity of different extraction methods.Result:The protein content of G.swinhonis was reduced by about 30%after fermentation,and the protein electrophoretic bands with molecular weight greater than 10 kDa disappeared.The anti-HepG2 tumor cell proliferation activity increased in vitro,and the IC 50 value of ultrasound extract induced HepG2 tumor cell apoptosis was 10.769 mg·mL-1,which was about 50%lower than that before fermentation.Conclusion:The growth and metabolism of Paecilomyces cicadae decomposed some proteins of G.swinhonis,and the activity of G.swinhonis against HepG2 tumor cells increased after fermentation by Paecilomyces cicadae.
