摘 要:目的:采用HPLC-ELSD,建立肉苁蓉药材中润肠通便有效组分甜菜碱、甘露醇、果糖、葡萄糖、蔗糖的多指标含量测定方法。方法:采用Shodex Ashaipak NH2P-504E色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(77∶23),流速为0.7 mL·min-1;柱温为25℃,检测器漂移管温度为100℃,气体流量为3.0 L·min-1。结果:在建立的液相色谱条件下,5个待测成分分离完全,甜菜碱、甘露醇、果糖、葡萄糖、蔗糖进样量分别在0.0992~2.9760、0.1988~5.9640、0.5108~6.1296、1.2176~6.5292、0.1984~5.9532μg线性良好;精密度、重复性、稳定性及回收率试验结果均符合含量测定要求,甜菜碱、甘露醇、果糖、葡萄糖和果糖的平均回收率(n=6)分别为98.49%、100.48%、99.27%、98.39%、98.97%。58批荒漠肉苁蓉样品中甜菜碱、甘露醇、果糖、葡萄糖和蔗糖的含量分别在2.73%~17.65%、2.46%~16.34%、2.52%~22.74%、0.97%~14.06%、3.15%~27.16%,10批管花肉苁蓉样品中甘露醇、果糖和葡萄糖的含量分别在1.00%~13.04%、2.98%~8.85%、1.09%~5.33%,未检出甜菜碱及蔗糖成分。结论:该方法简便、灵敏、准确性高、重复性好,能够实现对肉苁蓉药材润肠通便功效的质量控制,同时可用于区分荒漠肉苁蓉与管花肉苁蓉。甜菜碱和蔗糖含量的不同为肉苁蓉不同基原植物质地和生境的不同提供了理论依据。Objective:To develop an HPLC-ELSD method for simultaneous determination of laxative components,betaine,mannitol,fructose,glucose and sucrose in Cistanches Herba.Methods:The samples were separated on a Shodex Ashaipak NH2P-504E(250 mm×4.6 mm,5μm)column at 25℃.The mobile phase was acetonitrile-water(77∶23),and the flow rate was 0.7 mL·min-1.The detector drift tube temperature was 100℃,and the gas flow rate was 3.0 L·min-1.Results:The linear ranges for betaine,mannitol,fructose,glucose and sucrose were 0.0992-2.9760μg,0.1988-5.9640μg,0.5108-6.1296μg,1.2176-6.5292μg and 0.1984-5.9532μg,respectively;and their average recoveries were 98.49%,100.48%,99.27%,98.39%and 98.97%,respectively.The contents of betaine,mannitol,fructose,glucose and sucrose in 58 batches of C.deserticola were 2.73%-17.65%,2.46%-16.34%,2.52%-22.74%,0.97%-14.06%,and 3.15%-27.16%,respectively.While the contents of mannitol,fructose,and glucose in 10 batches of C.tubulosa were 1.00%-13.04%,2.98%-8.85%,and 1.09%-5.33%,respectively,and betaine and sucrose were undetected.Conclusions:The established HPLC quantitative method is simple,sensitive,accurate,and reproducible,and can be used to control the quality of Cistanches Herba and distinguish between C.deserticola and C.tubulosa.The different contents of betaine and sucrose supply a theoretical basis for the two different source plants of Cistanches Herba.
