摘 要:目的通过SD大鼠孕期、哺乳期镉暴露,探讨镉对成年雄性仔鼠生精功能的影响。方法SD大鼠受孕后,随机将其分为4个组,即对照组和低、中、高剂量染镉组。于SD大鼠受孕第9天起,对照组用生理盐水灌胃,低、中、高剂量组用氯化镉灌胃(剂量分别为10、20、40 Mg/kg·d^-1),直至哺乳期结束。待90日龄时,获取各组雄性仔鼠,断头处死,通过精子分析仪,观察睾丸每日精子生成量、精子活动度、附睾尾精子计数、精子畸形率;称取睾丸重量,计算脏器系数;通过原子吸收光谱法测定睾丸镉含量。结果与对照组比较,低、中、高剂量组每日精子生成量、精子活动率、附睾精子计数下降,精子畸形率升高(P<0.05),睾丸重量和脏器系数降低(P<0.05)。与低剂量组比较,中、高剂量组各项指标均有统计学差异(P<0.05),与中剂量组比较,高剂量组各项指标均有统计学差异(P<0.05)。结论SD大鼠孕期、哺乳期镉暴露,可降低子代雄性大鼠的精子质量,可能影响生育功能。Objective The effects of cadmium on spermatogenic function of adult male offspring of SD rats exposed to cadmium during pregnancy and lactation were studied.Methods After conception,SD rats were randomly divided into four groups:control group and low,middle and high dose cadmium groups.From the 9th day of conception of SD rats,the control group was given normal saline,and the low,middle and high dose groups were given cadmium chloride(10,20,40 mg/kg/d,respectively)until the end of lactation.At the age of 90 days,the male offspring of each group were decapitated and sacrificed.The daily sperm production,sperm motility,epididymal tail sperm count and sperm deformity rate were observed by sperm analyzer.The weight of testis was recorded and the organ coefficient was calculated.The content of cadmium in testis was determined by atomic absorption spectrometry(AAS).Results Compared with the control group,the daily sperm production,sperm motility,epididymal sperm count were decreased,sperm deformity rate increased(P<0.05),testicular weight and organ coefficient decreased(P<0.05).Compared with the low dose group,all the indexes in the middle dose group and high dose group were significantly different(P<0.05),and those in the high dose group were significantly different from those in the middle dose group(P<0.05).Conclusion Exposure to cadmium during pregnancy and lactation in SD rats can reduce the sperm quality of offspring male rats,it may affect the fertility function of male rats.
