摘 要:目的探讨荧光原位杂交技术(FISH)在快速鉴定石蜡包埋组织中的真菌感染的应用价值。方法制备针对曲霉菌属18SrRNA序列互补的寡核苷酸探针18S-1,经绿色荧光染料6-FAM标记后,对50例怀疑或证实为真菌感染的石蜡包埋标本进行FISH检测。结果 50例石蜡包埋标本中,HE染色、PAS染色、六胺银染色和真菌荧光染色显示真菌感染阳性例数分别为44、50、50和46例,32例FISH检测呈阳性反应。对照组为阴性。结论 FISH可以特异性地检测出石蜡包埋组织中的曲霉菌,是一种快速准确检测石蜡包埋组织内真菌感染的有效方法。Objective To investigate the application value of fluorescence in situ hybridization in rapid identification of fungal infection in paraffin-embedded tissues.Methods The oligonucleotide probe 18S-1 complementary to the 18SrRNA sequence of Aspergillus was prepared and labeled with the 6-FAM.50 paraffin-embedded specimens which suspected or confirmed to be fungal infection were subjected to FISH detection.Results Among the 50 paraffin-embedded specimens,HE staining,PAS staining,silver staining and fungal fluorescence staining were positive for 44 cases,50 cases,50 cases and 46 cases,respectively.Only 32 cases were positive by FISH.The control groups were negative.Conclusion Fluorescence in situ hybridization is a sensitive and specific method for rapid detection of Aspergillus infection in paraffin-embedded tissues.It is an effective method for rapid and accurate detection of fungal infections in paraffin-embedded tissues.
