左旋多巴结晶母液萃取物对抑制NG108-15细胞增殖作用的研究

摘　　要：本实验研究不同极性萃取溶剂制备的左旋多巴结晶母液萃取物体外抑制NG108-15细胞增殖的作用。在以猫豆为原料生产左旋多巴过程中,分别用石油醚、乙酸乙酯和正丁醇对结晶母液浓缩物进行萃取,然后采用四氮唑盐法(MTT法)检测各萃取物体外对NG108-15细胞增殖的抑制作用。实验结果显示,与空白组比较,石油醚、乙酸乙酯和正丁醇的萃取物对NG108-15细胞增殖均表现出明显的抑制作用(P<0.05或P<0.01),且抑制作用随着浓度的增加而增强;IC50分别为37.14μg·mL^-1、44.89μg·mL^-1、49.03μg·mL^-1。说明左旋多巴结晶母液对NG108-15肿瘤细胞的增殖有显著的抑制作用,提示母液中含有一些具有良好抑制肿瘤细胞活性的成分,有待进一步研究开发。The proliferation inhibition effect of levodopa crystallization mother liquor extracts prepared by different polar extraction solvents on NG108-15 cell in vitro was investigated.In the process of producing levodopa fromStizolobium cochinchinensis(Lour),petroleum ether,ethyl acetate and n-butanol were used to extract the crystallization mother liquor concentrate respectively,and then MTT assay was used to detect the inhibitory effect of each extract on the proliferation of NG108-15 cell.The experimental results showed that,compared with the blank group,the extracts of petroleum ether,ethyl acetate and n-butanol all showed a significant inhibitory effect on the proliferation of NG108-15 cell(P<0.05 or P<0.01),and the inhibitory effect became stronger while the extract concentrations increased.The IC50 was 37.14μg·mL^-1,44.89μg·mL^-1,49.03μg·mL^-1,respectively.It shows that the levodopa crystal mother liquor has a significant inhibitory effect on the proliferation of NG108-15 tumor cell,suggesting that the mother liquor contains some components that have good tumor cell inhibitory activity,which needs further research and development.
