摘  要:本研究旨在探索不同质量浓度的猪小肠抗菌肽对小鼠迟发型变态反应的影响和猪小肠抗菌肽的药理活性。选取36只体重在20 g左右的健康昆明小鼠,随机分成6组,分别为试验组1、试验组2、试验组3、试验组4、对照组和空白组,每组6只。试验组1、试验组2、试验组3、试验组4小鼠皮下注射多肽含量为0.1 mg/mL、0.3 mg/mL、0.5 mg/mL和原液0.7 mg/mL的猪小肠抗菌肽,注射剂量为0.01 mL/g;对照组及空白组分别皮下注射生理盐水,注射剂量为0.01 m L/g,连续注射7 d,之后将试验组和对照组采用二硝基氟苯(DNFB)致敏,剂量50μL/只,致敏5 d后,采用DNFB以剂量10μL/只进行二次攻击(空白组未致敏但有攻击),24 h后,以小鼠左右耳片重量之差(肿胀度)代表迟发型变态反应剧烈程度。结果显示,试验组1、2、3及4小鼠的肿胀度均值分别为9.4 mg、9.6 mg、10.1 mg和10.1 mg,均显著(P<0.05)高于对照组(7.8 mg)和空白组(0.7 mg)。结果表明,猪小肠抗菌肽对小鼠的迟发型变态反应具有增强作用,猪小肠抗菌液能够提高机体免疫水平。The purpose of this study was to explore the effects of different concentrations of porcine small intestine antibacterial peptides on delayed allergic reaction in mice and the pharmacological activity of porcine small intestinal antibacterial peptides.Thirty-six healthy Kunming mice weighing about 20 g were randomly divided into six groups,namely experimental group 1,experimental group2,experimental group 3,experimental group 4,control group and blank group,with 6 mice in each group.Experimental group 1,experimental group 2,experimental group 3,and experimental group 4 mice were injected subcutaneously with a peptide content of 0.1 mg/mL,0.3 mg/mL,0.5 mg/mL,and a stock solution of 0.7 mg/mL antibacterial peptides in pig small intestine 0.01 mL/g;the control group and the blank group were injected subcutaneously with saline at a dose of 0.01 mL/g for 7 consecutive days,and then the experimental group and the control group were sensitized with dinitrofluorobenzene(DNFB)50μL/head,after 5 days of sensitization,DNFB was used for a second challenge at a dose of 10μL/head(the blank group was not sensitized but challenged),and after 24 hours,it was represented by the difference between the left and right mouse ear weights(swelling)the severity of late-type allergies.The results showed that the average swelling degrees of the mice in the experimental group 1,2,3 and 4 were 9.4 mg,9.6 mg,10.1 mg and 10.1 mg,respectively,which were significantly higher(P<0.05)than those in the control group(7.8 mg)and the blank group(0.7 mg).The results showed that the pig small intestine antibacterial peptide had an enhanced effect on the delayed allergic reaction in mice,indicating that the pig small intestine antibacterial solution could improve the body’s immune level.
