摘 要:目的对比观察单次大剂量、多次小剂量环磷酰胺(CY)腹腔注射构建小鼠免疫抑制模型的效果。方法30只ICR健康雄性小鼠随机分为单次组、多次组、对照组各10只,单次组在实验第7天腹腔注射100 mg/kg的CY,多次组在实验第1、3、5、7 d腹腔注射50 mg/(kg·d)的CY,对照组在实验第1、3、5、7 d腹腔注射等量生理盐水。比较各组小鼠体质量、脾脏指数、胸腺指数、PPs计数、CD3^+细胞比例、CD19+细胞比例、肠道黏膜SIgA含量。结果与空白组比较,单词组实验第8 d及多次组实验第3、5、7、8 d体质量降低(P均<0. 05);与多次组比较,单次组实验第3、5、7、8 d体质量升高(P均<0. 05)。与空白组比较,单词组和多次组脾脏指数、胸腺指数降低(P均<0. 05);与多次组比较,单次组脾脏指数、胸腺指数降低(P均<0. 05)。与空白组比较,PPs计数、CD19^+细胞比例、肠道黏膜SIgA含量降低,CD3^+细胞比例升高(P均<0. 05);与多次组比较,单词组PPs计数、CD19^+细胞比例、肠道黏膜SIgA含量降低,CD3^+细胞比例升高(P均<0. 05)。结论单次大剂量和多次小剂量的CY均可建立免疫抑制动物模型,前者对小鼠体质量影响更小,且造模效果佳。Objective To compare the effects of single high dose and multiple low dose cyclophosphamide( CY) abdominal injection in establishing the appropriate immunosuppression mouse models. Methods Thirty healthy male ICR mice were randomly divided into the control group,single dose group,and multiple dose group,respectively. The mice in the single dose group were intraperitoneally injected with CY 100 mg/kg on the 7 th day,the mice in multiple dose group were intraperitoneally injected with CY 50 mg/( kg·d) on the 1 st,3 rd,5 th and 7 th days,while the mice in the control group were intraperitoneally injected with normal saline on the the 1 st,3 rd,5 th and 7 th days. The body weight,visceral index,thymus index,PPs count,the proportion of CD3^+,the proportion of CD19^+ and intestinal mucosa SIg A content of mice were compared between groups. Results Compared with the blank group,the body weight of the mice in the single dose group on the 8 th day and in the multiple dose group on the 3 rd,5 th,7 th and 8 th days decreased( all P < 0. 05).Compared with the blank group,the viscera index and thymus index decreased in the single dose group and multiple dose group( both P < 0. 05);compared with the multiple dose group,the viscera index and thymus index decreased in the single dose group( both P < 0. 05). Compared with the blank group,the intestinal mucosa PPs,proportion of CD19^+ and intestinal mucosa SIg A in the single dose group and multiple dose group decreased significantly,and the proportion of CD3^+ increased( all P < 0. 05). Compared with the multiple dose group,the intestinal mucosa PPs,proportion of CD19^+ and intestinal mucosa SIg A decreased significantly,and the proportion of CD3^+ increased in the single dose group( all P <0. 05). Conclusion Both the single high dose and multiple low doses of CY can establish animal models of immunosuppression,and the former has less influence on body weight and better modeling effect.
