摘 要:目的考察不同提取方式对金线莲多糖含量、结构及抗氧化活性的影响。方法采用热水回流提取法、超声提取法和酶法分别提取金线莲多糖,苯酚硫酸法测定多糖含量,刚果红实验检测三螺旋结构,采用DPPH自由基清除实验评价其抗氧化活性。结果不同提取方式对多糖的产率和DPPH清除活性均有影响,其中超声提取和酶解提取得到的多糖DPPH自由基清除活性优于热水回流提取所得金线莲多糖;提取所得金线莲多糖不含三螺旋结构。结论综合考虑提取效率、抗氧化活性和工艺成本,超声提取为最优方法。OBJECTIVE To investigate the effects of different extraction methods on the content,structure and antioxidant activity of polysaccharides from Anoectochilus roxburghii.METHODS Polysaccharide was extracted by hot water reflux extraction method,ultrasonic extraction method and enzymatic method.The polysaccharide content was determined by phenol sulfuric acid method.The three-helical structure was detected by Congo red experiment.The antioxidant activity was evaluated by DPPH free radical scavenging experiment.RESULTS Different extraction methods had effects on the yield of polysaccharides and DPPH scavenging activity.The DPPH free radical scavenging activity of polysaccharides extracted by ultrasonic extraction and enzymatic hydrolysis was better than that obtained by hot water reflux extraction.The gold obtained by different extraction methods Clematis polysaccharide does not contain a triple helix structure.CONCLUSION Considering extraction efficiency,antioxidant activity and process cost,ultrasonic extraction can be used to extract Astragalus polysaccharides.
