摘 要:旨在了解AraC家族转录调控因子lmo2672对单核增生李斯特菌(LM)生物被膜(BF)形成的影响。根据同源重组技术构建LM-△lmo2672基因缺失株,利用结晶紫染色法比较LM EGD-e和LM-△lmo2672在不同温度、渗透压、H 2O 2和胆酸盐环境中BF形成量的差异;通过qRT-PCR分析上述不同应激环境中BF相关基因相对转录水平的变化。研究结果证实:与LM EGD-e相比,LM-△lmo2672在4℃,30℃,5%NaCl,8%NaCl,5 mmol/L H 2O 2和0.3%胆酸盐培养环境中BF形成量显著降低;在37℃LM-△lmo2672 BF形成能力极显著降低;在不同应激环境中,LM-△lmo2672 BF形成相关基因的表达量均出现不同程度下降。研究结果表明,lmo2672基因在LM BF形成过程中发挥重要作用。In order to explore the function of AraC family regulator lmo2672 on the biofilm formation of LM,△lmo2672 deletion strain was constructed by using homologous recombination technique.We monitored their biofilm formation at different temperatures,osmotic pressures,media containing H 2O 2 and,bile salts we also explored the influences of lmo2672 deletion on the transcription of genes related to biofilm of LM using qRT-PCR.The results showed that compared with LM EGD-e,the biofilm biomass of lmo2672 deletion mutant strain significantly decreased under 4℃,30℃,5%NaCl,8%NaCl,5 mmol/L H2O2 and 0.3%bile salts stresses,and biofilm formation of the lmo2672 deletion mutant was significantly compromised under 37℃.Furthermore,in different environmental stresses,the expressions of the biofilm-related genes decreased to different degrees in LM-Δlmo2672.Therefore,these findings suggested that the lmo2672 of AraC family plays important roles in the Listeria monocytogenes biofilm formation.
