摘 要:以东亚钳蝎毒素和梁氏异蝎毒素来研究蝎毒对Hela细胞增殖的影响,MTT法试验结果:20,40,60μg/m L的2种蝎毒对Hela细胞均有抑制增殖的作用,具有浓度和时间的依赖关系,随着浓度升高和时间的延长,Hela细胞增殖的抑制作用增强。淋巴细胞转化试验结果:20,40,60μg/m L的2种蝎毒均能促进淋巴细胞转化,并且梁氏异蝎毒素的作用更强。乳酸脱氢酶(LDH)释放试验结果:2种毒素均能促使LDH的释放,随着蝎毒浓度的增加和时间的延长,Hela细胞溶液上清液中LDH释放量逐渐增加。由此证明,这2种蝎毒均可抑制Hela细胞增殖,有望成为一种抗肿瘤药物。The effect of Buthus martensii Karsch and Heterometrus liangi venom on the proliferation of Hela cells was studied.The results of MTT assay showed that 20,40,60|xg/mL of two scorpion venom inhibited the proliferation of Hela cells and there was relationship between concentration and time dependent as the concentration increased and the time prolonged,the inhibition to Hela cell proliferation was enhanced.The results of lymphocyte transformation experiment showed that 20,40,60|xg/mL of two scorpion venom could promote lymphocyte transformation,and the effect of Heterometrus liangi venom was stronger than that of Buthus martensii Karsch.The results of lactate dehy・drogenase(LDH)release experiment showed that both venoms could promote the release of LDH,and with the increase of scorpion venom concentration and time prolonging,the release of LDH in the supernatant of Hela cells gradually increased.It was proved that these two kinds of scorpion ven・oms could inhibit the proliferation of Hela cells and were expected to be develop an anti-tumor drug.
