摘 要:目的评价香参丸治疗实验性结肠炎大鼠疗效,并从香参丸对结肠炎大鼠结肠黏膜关键酶的调节作用探讨其作用机制。方法将40只SD大鼠随机平均分成正常组、模型组、香参丸组、美沙拉嗪组,给予3mg/kg TNBS/50%乙醇混合溶液灌肠复制模型,灌胃给药7d。通过测量结肠重量和长度、计算结肠重量指数、观察组织病理切片评价香参丸疗效,检测结肠黏膜ADH、ChAT、NADP^+、ACh和MAO含量。结果与模型组比较,香参丸组结肠长度明显增长,重量和结肠重量指数下降,ADH、ChAT和NADP^+均下降,而ACh和MAO显著升高(P<0.01或P<0.05)。结论香参丸可能通过下调结肠黏膜ADH和NADP^+,上调Ach的表达量起到治疗实验性大鼠结肠炎作用。Objective To evaluate the efficacy of Xiangshenwan in treatment of experimental colitis in rats and to explore the mechanism of action of Xiangshenwan on the key enzymes in colonic mucosa of colitis rats.Methods Forty SD rats were randomly divided into normal group, model group, Xiangshenwan group, and mesalazine group. The rats were given a 3 mg/kg TNBS/50% ethanol enema model, intragastrically administered for 7 days, and sacrificed on the 8 th day. By measuring the weight and length of the colon, calculating the colon index, and observing the histopathological sections to evaluate the efficacy of the drug, the ADH, ChAT, NADP^+, ACh, and MAO levels in the colonic mucosa were measured.Results Compared with the model group, the colon length, weight and colon index of the Xiangshenwan group decreased significantly, ADH, ChAT and NADP^+ decreased, while ACh and MAO increased significantly(P<0.01 or P<0.05). Conclusion Xiangshenwan is possible to treat experimental colitis in rats by down-regulating ADH and NADP^+ in the colonic mucosa and up-regulating the expression of Ach.
