摘 要:目的研究羌活对佐剂关节炎大鼠背根神经节P2X受体表达的影响,探讨羌活的镇痛作用机制。方法建立弗氏完全佐剂关节炎大鼠模型。采用机械压力刺激法检测大鼠足趾机械痛阈值,ELISA法检测大鼠足趾肌肉组织ATP含量,并通过RT-PCR检测大鼠背根神经节中P2X3、P2X4、P2X6受体mRNA的表达情况。结果羌活能显著升高关节炎大鼠机械压力痛阈值(P<0.01),降低ATP含量(P<0.05,并能明显下调背根神经节中P2X3、P2X4、P2X6受体mRNA的表达(P<0.05)。结论羌活对佐剂关节炎大鼠具有镇痛作用,其作用机制可能与抑制背根神经节中P2X3、P2X4、P2X6受体的表达有关。Objective To investigate the effect of Notopterygium incisum on the expression of P2 X receptor in dorsal root ganglia of adjuvant arthritis rats, and to elucidate the analgesic mechanism of Notopterygium incisum. Methods Rats were established into complete Freund’s adjuvant arthritis rats model. Then mechanical pressure stimulation was used to measure mechanical pain threshold, and ELISA for ATP content. After that, RT-PCR was utilized to detected the expression of P2 X3, P2 X4 and P2 X6 mRNA in rats dorsal root ganglia.Results Notopterygium incisum could remarkable increase the mechanical pain threshold(P<0.01) and decrease ATP content(P<0.05) in arthritic rats;also significantly down-regulate the expression of P2 X3, P2 X4 and P2 X6 mRNA(P<0.05). Conclusion Notopterygium incisum show the analgesic effect on adjuvant arthritis rats, which might be due to its inhibition the expression of P2 X3, P2 X4 and P2 X6 mRNA in the dorsal root ganglia.
