摘  要:目的研究四妙丸对尿酸钠晶体(MSU)诱导的巨噬细胞表达白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的影响,并从调节核因子κB(NF-κB)活化途径探讨其机制。方法四妙丸与MSU共同作用于RAW264.7细胞24 h。MTT法检测细胞活力,酶联免疫吸附法检测细胞培养液中IL-1β和TNF-α含量,Western blot法分析细胞内NF-κB抑制蛋白(IκBα)含量,同时分析NF-κB与DNA的结合活性。结果四妙丸显著降低了MSU刺激的RAW264.7细胞培养液中IL-1β和TNF-α含量(P<0.05或P<0.01);与此同时,四妙丸还明显提高了MSU刺激的RAW264.7细胞内IκBα含量(P<0.05或P<0.01),并使NF-κB与DNA的结合活性明显降低(P<0.05或P<0.01)。结论四妙丸可通过抑制MSU诱导的巨噬细胞转录因子NF-κB的活化而降低促炎介质IL-1β和TNF-α的表达。Objective To investigate the effects of Simiao Wan on pro-inflammatory mediator production and nuclear factor NF-κB activation in monosodium urate(MSU)-induced RAW264.7 cells.Methods RAW264.7 cells were treated with Simiao Wan and MSU for 24 h. Cell viability was tested by MTT assay. Levels of interleukin IL-1β and tumor necrosis factor TNF-α in culture supernatant were measured by enzyme-linked immunosorbent assay. Level of inhibitor of NF-κB(IκBα) was evaluated by Western blot assay. DNA binding activity of NF-κB was evaluated by Trans AMTM kit for NF-κB p65. Results Simiao Wan significantly decreased the levels of IL-1β and TNF-α in culture supernatant of MSU-induced RAW264.7 cells(P<0.05 or P<0.01). Meanwhile, Simiao Wan significantly enhanced IκBα content(P<0.05 or P<0.01), and lowered DNA binding activity of NF-κB in MSU-induced RAW264.7 cells(P<0.05 or P<0.01). Conclusion Simiao Wan decreased the pro-inflammatory mediator production through inhibiting the NF-κB activation in MSU-induced RAW264.7 cells.
