摘 要:目的研究越鞠甘麦大枣汤(YG)对脂多糖(LPS)诱导的BV2细胞损伤的保护作用。方法用LPS溶液建立BV2细胞损伤模型,制备YG无菌溶液,采用四甲基偶氮唑盐(MTT)法检测各组细胞成活率,ELISA法测定各组肿瘤坏死因子-α(TNF-α),白细胞介素-1β(IL-1β),白细胞介素-10(IL-10)的含量,蛋白免疫印迹法(Western blot)法测定各组细胞内核因子-κB(NF-κB)p65,蛋白激酶B(AKT),磷酸化蛋白激酶B(p-AKT)蛋白表达水平。结果模型组细胞成活率下降,细胞外液中TNF-α,IL-1β的含量上升,而IL-10的含量没有显著变化;细胞内AKT,p-AKT,NF-κB(p65)蛋白表达水平提高;YG能提高LPS模型条件下的细胞成活率,使细胞外液中TNF-α,IL-1β,IL-10的含量均下降,细胞内AKT,p-AKT,NF-κB(p65)蛋白表达水平下降。结论 YG对LPS诱导的BV2细胞损伤具有保护作用。Objective To study the protective effect of Yueju Ganmai Dazao Decoction(YG) on lipopolysaccharide(LPS)-induced BV2 cell injury.Methods The injury model of BV2 cells was established by LPS solution. The YG sterile solution was prepared. The survival rate of each group was detected by MTT assay. The levels of cytokines TNF-α, IL-1β and IL-10 in each group were determined by ELISA. Western blot was used to determine the expression levels of AKT, p-AKT and NF-KB(p65) in each group. Results The survival rate of cells in the model group decreased, the levels of TNF-α and IL-1β in the extracellular fluid increased, the IL-10 content did not change significantly;the expression levels of AKT, p-AKT and NF-κB(p65) in the cells increased. YG can increase the survival rate of cells under LPS model, decrease the content of TNF-α, IL-1β and IL-10 in extracellular fluid, and decrease the expression of AKT, p-AKT and NF-κB(p65) in cells.Conclusion YG has a protective effect on LPS-induced BV2 cell injury.
