摘 要:目的观察不同时期电针少阳经特定穴对偏头痛大鼠的行为学、50%缩足阈值(50%PWT)及血清中5-羟色胺(5-HT),P物质(SP)含量的影响,为临床运用针灸治疗偏头痛选择合理方案。方法将50只大鼠随机分为空白组(A组)、模型组(B组)、电针少阳经穴预防组(C组)、电针少阳经穴治疗组(D组)、电针少阳经穴预防+治疗组(E组)共5组,每组10只。C组和E组捆绑行电针治疗30 min×7天,其余各组同时段捆绑30 min,7天。A组第8天皮下注射等量0.9%氯化钠注射液,30 min后捆绑30 min;其余各组第8天皮下注射硝酸甘油造模,其中B组、C组造模30 min后捆绑30 min,D、E组造模30 min后捆绑行电针治疗30 min。观察造模前后及治疗后大鼠行为学,通过纤维丝疼痛测试仪测量各组大鼠造模前后及治疗后的50%PWT,采用酶联免疫吸附测定法(ELISA)中测定血清5-HT,SP的含量。结果与A组比较,造模后B组大鼠行为学评分明显升高(P<0.01),50%PWT明显降低(P<0.01);与B组比较,电针少阳经穴各组(CDE三组)治疗后行为学评分明显降低(P<0.01),50%PWT明显升高(P<0.01),血清5-HT含量升高,SP含量降低(P<0.01);分别与C组、D组比较,E组治疗后行为学评分明显降低(P<0.01),50%PWT明显升高(P<0.01),血清5-HT含量显著升高,SP含量明显降低(P<0.01)。结论电针少阳经特定穴对偏头痛大鼠有明显的镇痛作用,同时电针少阳经特定穴预防+治疗改善偏头痛大鼠疼痛程度的效果优于单纯预防性治疗和单纯发作期治疗。Objective To observe the effect of electroacupuncture on the behavior of migraine rats,50% Paw Withdrawal Threshold(PWT)and the content of 5-hydroxy tryptamine(5-HT)and Substance p(SP)in serum in different periods, and to select a reasonable plan for the clinical use of acupuncture for the treatment of migraine.Methods Fifty rats were randomly divided into five groups(group A), model group(group B), electroacupuncture Shaoyang acupoint prevention group(group C), electroacupuncture Shaoyang acupoint treatment group(group D) and electroacupuncture Shaoyang acupoint prevention and treatment group(group E), with 10 rats in each group. Patients in group C and group E were treated with electroacupuncture for 30 min x 7 days, while patients in other groups were treated for 30 min x 7 days at the same time. Group A was subcutaneously injected with the same 0.9% sodium chloride injection on the 8 th day, which was bundled for 30 minutes after 30 minutes. The other groups were subcutaneously injected with nitroglycerin at the eighth day, in which group B and group C were bound for 30 min, and group D and E were bound for 30 min. To observe the behavior of rats before and after model building and after treatment, 50%PWT before and after model building and after treatment were measured by fibrosis pain tester, and serum 5-HT and SP contents were measured by ELISA.Results Compared with group A, the behavioral scores of rats in group B after model building increased significantly(P < 0.01), and 50% PWT decreased significantly(P < 0.01). Compared with group B, the behavioral score of each group(the three groups of CDE) at the acupoint of Shaoyang electroacupuncture was significantly reduced after treatment(P < 0.01), 50% PWT was significantly increased(P < 0.01), serum 5-ht content was increased, and SP content was reduced(P < 0.01). Compared with group C and group D, the behavioral score of group E was significantly reduced(P < 0.01), 50% PWT was significantly increased(P < 0.01), serum 5-ht content was significantly increased
