摘  要:目的探讨五味沉香散对慢性低氧大鼠海马神经细胞损伤的保护作用。方法将大鼠分为低氧对照组(生理盐水组)、阳性对照组(红景天组)、低氧药物组(五味沉香散组),每组10只,均置海拔5500 m低氧环境30 d时取脑组织。通过免疫组化及酶联免疫吸附实验法对脑组织行HIF-1α、Beclin-1、Bcl-2水平测定,同时制作脑组织形态学切片。对相关检测和观察结果作相关性分析。结果五味沉香散组和红景天组HIF-1α、Beclin-1水平显著低于生理盐水组(P<0.05);五味沉香散组和红景天组Bcl-2水平显著高于生理盐水组(P<0.05)。通过免疫组化方法发现:三组大鼠脑组织HIF-1α、Beclin-1、Bcl-2水平与酶联免疫吸附实验结果一致。结论五味沉香散对慢性低氧大鼠海马神经细胞损伤有修复和保护作用。Objective To investigate the protective effect of Wuweichenxiangsan on hippocampal nerve cell injury in chronic hypoxia rats.Methods A total of 30 rats were evenly divided into hypoxic control group(normal saline group),positive control group(Rhodiola rosea group),and hypoxic drug group(Wuweichenxiangsan group).Brain tissues were taken at 30 d in a hypoxic environment at an altitude of 5500 m.HIF-1α,Beclin-1 and Bcl-2 were measured in brain tissues by immunohistochemistry and enzyme-linked immunosorbent assay and brain tissues morphological sections were made.Correlation analysis was made between detection and observation results.Results The levels of HIF-1αand Beclin-1 in both Wuweichenxiangsan and Rhodiola groups were significantly lower than those in the normal saline group(P<0.05);the Bcl-2 levels in both Wuweichenxiangsan and Rhodiola groups were predominantly higher than those in the normal saline group(P<0.05).It was found that the levels of HIF-1α,Beclin-1 and Bcl-2 in the brain tissues by immunohistochemistry methods were consistent with the results of enzyme-linked immunosorbent assay of the three groups of rats.Conclusion Wuweichenxiangsan can repair and protect hippocampal nerve cells damage from chronic hypoxia in rats.
