摘 要:本研究采用玉米幼胚为受体材料,通过对15种不同玉米自交系幼胚进行植物组织培养,筛选出具有最高愈伤诱导率的玉米自交系SL1303,其愈伤诱导率为93%,且愈伤分化再生能力最强。通过取用授粉后不同时期的玉米幼胚进行组织培养,发现当幼胚大小在1~1.5 mm时愈伤诱导率最高。进一步将玉米PIFs转录因子ZmPIF3基因构建到植物表达载体P1011上,将该表达载体导入到农杆菌EHA105中,利用农杆菌介导法侵染玉米自交系SL1303的愈伤,诱导分化成再生植株。经PCR鉴定结合测序检测,共鉴定出12株阳性植株,获得了ZmPIF3基因在玉米中过表达的T0代植株,对进一步研究玉米中ZmPIF3基因的功能具有重要意义。In this study,maize immature embryos were used as acceptor materials.The maize inbred line SL1303 with the highest callus induction rate was obtained by screening 15 different maize inbred lines.The callus induction rate of SL1303 was 93%,and SL1303 had the highest callus differentiation rate.Plant tissue culture was carried out by using maize young embryos at different stages after pollination,the results showed that when the size of immature embryos was 1~1.5 mm,the callus induction rate was the highest.The maize PIFs transcription factor ZmPIF3 was constructed into expression vector P1011,which was introduced into Agrobacterium EHA105.The callus of SL1303 was infected with EHA105 and induced to differentiate into regenerated plants.A total of 12 positive plants were identified by PCR and sequencing.ZmPIF3 transgenic maize is important for further study of the function of the ZmPIF3 gene in maize.
