摘  要:本研究通过对中国樱桃优良品种‘黑珍珠’离体培养技术进行探索,解决了‘黑珍珠’组织培养中极易褐化的问题,建立了适宜的离体培养体系。本研究以当年生绿枝带芽茎段为外植体,探索了外植体最佳采集时间以及不同激素配比对初代培养、增殖继代培养和生根的影响等。结果表明,适宜的中国樱桃‘黑珍珠’离体培养体系为:初春(3月)取材,并用0.05%HgCl2消毒处理3 min,能有效控制初代培养中的污染和褐化问题。选择最适初代培养基为MS+0.8 mg/L 6-BA+0.3 mg/L IBA,诱导率为68.75%,最佳的继代增殖培养基为MS+1.0 mg/L 6-BA+0.4 mg/L IBA,增殖系数可达4.14%,生根阶段以1/2MS+0.2 mg/L 6-BA+0.4 mg/L IBA的培养基效果最佳,生根率85.71%,每株苗可生根5.5条,平均根长4.77 cm。本研究结果为其他樱属乃至木本果树植物的组织培养提供了较为可靠的参考,为易褐化的离体培养材料提供解决思路。同时,也为中国樱桃的优异性状基因发掘及遗传转化提供重要技术依据。In this study,we tried to establish an in vitro rapid propagation system of Chinese cherry'Heizhenzhu'and solve the brown problem.We explored the optimal collection time of explants and the effects of different hormone ratios on primary culture,subculture and rooting by using green shoots with bud stems as explants.The results showed that the approximate period materials were selected at early march.Materials should be disinfected with 0.05%of HgCl2 for 3 minutes to avoid the pollution and browning problems.The suitable culture system was MS+0.8 mg/L 6-BA+0.3 mg/L IBA,which reached 68.75%of explant induction rate.The best subculture medium was MS+1.0 mg/L 6-BA+0.4 mg/L IBA with 4.14%of proliferation coefficient.For rooting culture,the system was 1/2MS+0.2 mg/L 6-BA+0.4 mg/L IBA,which obtained 85.71%of rooting rate and 5.5 seedlings per seedling with average root length of 4.77 cm.These results not only provide a reliable reference for the tissue culture of other Cerasus Mill.plants,even woody fruit trees,and solutions for the easy-browning in vitro culture materials,but also lay the foundation for identifying and genetically transform the excellent traits of Chinese cherry.
