摘 要:【目的】为了研究不同浓度的丹参提取液对小鼠抗大肠杆菌感染的免疫活性调控作用。【方法】以灌胃方式对昆明小鼠给予不同浓度的丹参提取液,设丹参提取液的药物为0.0195、0.0390、0.0780g/mL灌胃饲养28d,观察不同浓度的丹参提取液对小鼠增重及大肠杆菌感染后血清细胞因子、免疫球蛋白和生化指标的影响。【结果】丹参提取液可促进小鼠机体的生长,显著降低大肠杆菌致死率,经丹参提取液预防干预的小鼠LD50降低至30%,对大肠杆菌攻毒后小鼠血清促炎因子IL-6、TNF-α有抑制作用,且以0.0195g/mL的丹参提取液对TNF-α的抑制效果最佳。0.0780g/mL的丹参提取液可极显著提高小鼠血清IgG浓度,且经丹参提取液预防干预的试验组血清生化肝功能指标极显著低于大肠杆菌模型组,与对照组无显著差异。【结论】丹参提取液能有效增强大肠杆菌诱导的小鼠肠道炎症的免疫调节作用,保护机体由大肠杆菌释放的脂多糖引起的肝损伤,减少炎症对机体的损害作用,提高小鼠的免疫力,发挥抗炎作用,可用于预防或治疗。【Objective】In order to study the effects of different concentrations of S.miltiorrhiza extract on the immune activity of mice against E.coli infection.【Methods】Different doses of S.miltiorrhiza extract were administered to Kunming mice by intragastric administration.The administration concentration of S.miltiorrhiza extract was 0.0195,0.0390,0.0780 g/mL,respectively,and lasted for 28 days.The effects of different concentrations of S.miltiorrhiza extract on the weight gain and serum cytokines,immunoglobulins and biochemical markers of mice after E.coli infection were observed.【Results】S.miltiorrhiza extract promoted the growth of mouse significantly,reduced the lethal rate and inhibited the pro-inflammatory factors in mice after E.coli challenge,the LD50 of mice treated with S.miltiorrhizaextract was reduced to 30%.In addition,the inhibitory effect of S.miltiorrhiza extract at the concentration of 0.0195 g/mL on TNF-αwas the best.S.miltiorrhiza extract at a concentration of 0.0780 g/mL significantly increased serum IgG concentration in mice,and its extract also protected liver caused by lipopolysaccharide released by E.coli.【conclusion】S.miltiorrhiza extract effectively regulates the immunoe of E.coli-induced intestinal inflammation in mice,protects liver caused by lipopolysaccharide released by E.coli,reduces the damage of inflammation to the body,improves the immunity of mice,and exerts anti-inflammation.It can be used for prevention or treatment.
