摘 要:【目的】通过测定不同产地板蓝根药材中(R,S)-告依春的含量,并分析产地对其含量的影响,为合理选择板蓝根种植区域提供参考。【方法】采用高效液相色谱法,选择Agilent HC-C18 (2)色谱柱,以甲醇-0.02%磷酸溶液为流动相,流速为1.0mL/min,检测波长为245nm,对5个不同产地板蓝根中(R,S)-告依春含量进行测量。【结果】方法学考察发现其线性范围、精密度、稳定性、重复性、加样回收率均符合试验规定。5个产地共10批板蓝根药材中的(R,S)-告依春含量范围为0.197~0.590mg/g,其中河北和山东产地的板蓝根中(R,S)-告依春含量较高,分别为0.590mg/g和0.481mg/g,然后依次为甘肃、东北和安徽产地。比较各生态因子,河北和山东地区的年平均气温和年降水量较适宜板蓝根中(R,S)-告依春的积累。【结论】不同产地板蓝根中(R,S)-告依春含量差异较大,其中河北和山东产地含量较高,气温和降水是影响其含量的主要生态因子。【Objective】By measuring(R,S)-goitrin in different production of Isatidis Radix,and analyzing the influence of the origin on its content,to provide a reference for rational selection of the planting area of Isatidis Radix.【Method】(R,S)-goitrin in Isatidis Radix from five different origins was determined by High Performance Liquid Chromatography(HPLC),Agilent HC-C18(2)column with methanol-0.02% phosphoric acid solution as mobile phase,flow rate at 1.0 mL/min,detection wavelength of 245 nm.【Results】In the methodological investigation,the linear range,precision,stability,repeatability,and sample recovery rate all meet the test requirements.The(R,S)-goitrin content in 10 batches of Isatidis Radix in the five producing areas ranged from 0.197 to 0.590 mg/g,and the highest content of(R,S)-goitrin in Isatidis Radix is Hebei and Shandong,0.590 mg/g and 0.481 mg/g,respectively,followed by Gansu,Dongbei and Anhui.Comparing the various ecological factors,the annual average temperature and annual precipitation in Hebei and Shandong are most suitable for the accumulation of(R,S)-goitrin in Isatidis Radix.【Conclusion】The content of Isatidis Radix varies greatly from different producing areas,of which Hebei and Shandong have the highest content,and temperature and precipitation are the main ecological factors affecting their content.
