摘 要:【目的】为了揭示中江县丹参连作土壤的细菌多样性。【方法】在中江丹参GMP基地采取患病丹参根际与非根际土壤、正常生长丹参根际与非根际土壤,采用可培养和免培养技术细菌研究供试土壤的细菌类群变化。【结果】应用稀释分离法,从供试土壤中获得46株可培养细菌,16S rDNA-RFLP分析表明,在79%相似水平分为7个遗传群,对代表菌株16S rDNA进行序列分析,构建了系统发育树,供试菌株为芽孢杆菌属(Bacillus)、短杆菌属(Brevibacterium)、拜叶林克氏菌属(Beijerinckia)和假单胞菌属(Pseudomonas)。【结论】PCR-DGGE图谱聚类分析表明,丹参连作显著影响了土壤细菌类群,供试土壤细菌类群间存在着明显的遗传多样性;条带克隆测序表明绝大多数菌株属于未培养细菌。【Objective】The study aimed to reveal soil bacteria diversity of Salvia miltiorrhiza continuous cropping soil in Zhongjiang county of Sichuan province.【Method】The rhizospheric and non-rhizospheric soil samples of both sick S.miltiorrhiza Bge and normal S.miltiorrhiza Bge were picked,and soil bacterial communities were analyzed by dilution plate method and PCR-DGGE.【Result】The results showed that a total of 46 bacterial strains were isolated and purified,and 16S rDNA PCR-RFLP analysis showed that there were seven genetic groups formed at the similarity of 79%.16S rDNA sequencing analysis of the representative bacterial isolates showed that these strains belonged to genus Bacillus,Brevibacterium,Beijerinckia and Pseudomonas.【Conclusion】PCR-DGGE analysis suggested that S.miltiorrhizae continuous cropping changed soil bacterial groups significantly;The genetic diversity of the bacteria groups existed among the tested soil samples;And the sequences of cloned PCR-DGGE bands indicated that most strains were uncultured bacteria.
