摘 要:【目的】为建立高效的火龙果快繁体系及保存母本优良性状。【方法】以成年火龙果植株茎段作外植体,研究不同生长调节剂及配比、基本培养基浓度、外植体类型等对茎段不定芽诱导的影响,筛选适宜不定芽增殖、生根及移栽等的最佳培养条件。【结果】火龙果茎段上保留刺座并带小刺和绒毛的外植体,其不定芽诱导率最高(85.1%),显著高于带刺座但未保留小刺和绒毛的外植体,而不带刺座的外植体不定芽诱导率为0;适宜火龙果不定芽诱导的培养基为MS+6-BA 2.0 mg/L+NAA 0.1~0.2 mg/L;不定芽增殖系数最高的培养基为MS+6-BA 0.5 mg/L+NAA 0.2 mg/L+Tryptone 500 mg/L;适宜不定芽生根的培养基为6-BA 1.0 mg/L+NAA 0.1~0.2 mg/L+CCC 0.5~1.0 mg/L+AC 0.03%~0.05%,生根率在98%以上;移栽成活率高、长势较好的基质是珍珠岩+腐殖土(2∶1)或蛭石+腐殖土(1∶1)。【结论】利用带完整刺座(包括小刺和绒毛)的火龙果成年植株茎段直接诱导不定芽的成功率较高,培养周期较短,生根移栽较易,为火龙果的工厂化育苗及快速推广应用奠定基础。【Objective】This paper aimed to establish an efficient pitaya fruit rapid propagation system and preserve the superior traits of the female parent.【Method】The stem segments of adult pitaya plant were used as explants to study the effects of different growth regulators and ratios,basic medium concentration and explant type on the induction of adventitious buds in stems.And thereby screen the best culture conditions suitable for adventitious buds proliferation,rooting and transplanting.【Result】The explants with splinters and nap on the stem seg-ments of pitaya had the greatest adventitious bud induction rate(85.1%),which was significantly greater than the explants with splinters but no thorns and fluff.The induction rate of adventitious buds of explants with no splinters was 0;The medium suitable for adventitious bud induction of pitaya was MS+6-BA 2.0 mg/L+NAA 0.1-0.2 mg/L;the medium with the greatest adventitious bud proliferation coefficient was MS+6-BA 0.5 mg/L+NAA 0.2 mg/L+Tryptone 500 mg/L;Suitable medium for adventitious bud rooting was 6-BA 1.0 mg/L+NAA 0.1-0.2 mg/L+CCC 0.5-1.0 mg/L+AC 0.03%-0.05%,the rooting rate was above 98%;The substrate with high survival rate and good growth rate was perlite+humus soil(2∶1)or vermiculite+humus soil(1∶1).
